Efek Ekstrak Kayu Manis (Cinnamomum burmannii) Terhadap Profil Histopatologi Tubulus Proksimal dan Jumlah Sel Limfosit pada Ginjal Tikus yang Diinduksi Streptozotocin

Kerusakan ginjal dan inflamasi kronik dapat terjadi akibat hiperglikemia persisten pada Diabetic Nephropathy. Kayu manis (Cinnamomum burmannii) secara empiris diketahui memiliki efek antihiperglikemik dan antioksidan sehingga berpotensi melindungi jaringan ginjal. Penelitian ini bertujuan menganalisis pengaruh ekstrak kayu manis terhadap gambaran histopatologi tubulus proksimal dan jumlah sel limfosit ginjal tikus yang diinduksi Streptozotocin (STZ). Desain yang digunakan adalah post-test only control group pada tikus Wistar jantan yang dibagi menjadi enam kelompok: kontrol sehat, STZ 35 mg/kgBB, STZ + metformin 9 mg/kgBB, serta STZ + ekstrak kayu manis dosis 150, 300, dan 600 mg/kgBB selama 21 hari. Preparat ginjal diamati menggunakan pewarnaan Hematoxylin and eosin stain. Analisis data dilakukan dengan uji Shapiro–Wilk, one-way ANOVA dan post hoc Tukey, sedangkan jumlah limfosit dianalisis menggunakan Kruskal–Wallis dan Mann–Whitney. Hasil menunjukkan kelompok STZ mengalami kerusakan tubulus proksimal berat dan peningkatan limfosit. Perbedaan signifikan ditemukan antara kelompok STZ dengan kelompok sehat, metformin, serta dosis 150 dan 300 mg/kgBB, namun tidak pada dosis 600 mg/kgBB. Dosis 150 mg/kgBB paling efektif dalam memperbaiki struktur tubulus dan menurunkan infiltrasi limfosit.

Kidney damage and chronic inflammation can occur as a result of persistent hyperglycemia in diabetic nephropathy. Cinnamon (Cinnamomum burmannii) is empirically known to have antihyperglycemic and antioxidant effects, thus potentially protecting kidney tissue. This study aims to analyze the effect of cinnamon extract on the histopathological picture of the proximal tubules and the number of kidney lymphocytes in rats induced by Streptozotocin (STZ). The design used was a post-test only control group in male Wistar rats divided into six groups: healthy control, STZ 35 mg/kgBW, STZ + metformin 9 mg/kgBW, and STZ + cinnamon extract doses of 150, 300, and 600 mg/kgBW for 21 days. Kidney preparations were observed using Hematoxylin and eosin stain. Data analysis was performed using the Shapiro–Wilk test, one-way ANOVA and post hoc Tukey, while lymphocyte counts were analyzed using Kruskal–Wallis and Mann–Whitney. The results showed that the STZ group experienced severe proximal tubule damage and an increase in lymphocytes. Significant differences were found between the STZ group and the healthy group, the metformin group, and the 150 and 300 mg/kgBW groups, but not in the 600 mg/kgBW group. The 150 mg/kgBW dose was most effective in improving tubule structure and reducing lymphocyte infiltration.
