Potensi Trichoderma atroviride dalam Mengendalikan Penyakit Karat Daun Kopi

Karat daun kopi yang disebabkan Hemileia vastatrix dapat menurunkan produksi kopi mencapai 35%. Pengendalian secara hayati perlu dilakukan agar produktivitas meningkat tanpa risiko pencemaran terhadap hasil produk dan lingkungan. Penelitian ini bertujuan untuk mengetahui kemampuan Trichoderma atroviride dalam menghambat H. vastatrix menggunakan T. atroviride dalam formula padat. Penelitian dilaksanakan Agustus 2024-April 2025 di Fakultas Pertanian Unsoed. Pengujian meliputi aktivitas enzim kitinase, protease, dan glukanase, uji pertumbuhan T. atroviride pada media padat beras, jagung, dan dedak; uji penghambatan perkecambahan H. vastatrix dan pengaplikasian formula padat T. atroviride. Pengujian formula aplikasi dilakukan dengan 6 ulangan dan perlakuan terdiri dari kontrol, formula padat T. atroviride konsentrasi 10 g/L, 25 g/L, dan 50 g/L. Variabel yang diamati meliputi intensitas penyakit, laju infeksi, AUDPC, dan kemampuan kolonisasi T. atroviride terhadap H. vastatrix. Hasil penelitian menunjukkan T. atroviride menghasilkan enzim pendegradasi dinding sel yaitu kitinase, protease, dan glukanase, T. atroviride mampu menghambat perkecambahan H. vastatrix sebesar 42%. Aplikasi formula padat T. atroviride konsentrasi 25 g/L dan 50 g/L mampu menurunkan perkembangan penyakit masing-masing sebesar 46,93% dan 48,93% melalui parasitisme dan produksi enzim pendegradasi dinding sel.

Coffee leaf rust caused by Hemileia vastatrix can reduce coffee production by up to 35%. Biological control is needed to increase productivity without the risk of contamination of products and the environment. This study aims to determine the ability of Trichoderma atroviride to inhibit H. vastatrix using T. atroviride in a solid formula. The study was conducted from August 2024 to April 2025 at the Faculty of Agriculture, Unsoed. Tests include the activity of chitinase, protease, and glucanase enzymes, growth tests of T. atroviride on solid media of rice, corn, and bran; germination inhibition test of H. vastatrix and application of solid formula of T. atroviride. The application formula test was carried out with 6 replications and the treatments consisted of control, solid formula of T. atroviride with concentrations of 10 g/L, 25 g/L, and 50 g/L. The variables observed included disease intensity, infection rate, AUDPC, and colonization ability of T. atroviride against H. vastatrix. The results showed that T. atroviride produces cell wall degrading enzymes, namely chitinase, protease, and glucanase, T. atroviride is able to inhibit the germination of H. vastatrix by 42%. Application of solid formula T. atroviride concentrations of 25 g/L and 50 g/L can reduce the development of the disease by 46.93% and 48.93% through parasitism and production of cell wall degrading enzymes.
