Infeksi Staphylococcus aureus, khususnya strain MRSA, menjadi masalah serius karena resistensinya terhadap berbagai antibiotik. Daun saga (Abrus precatorius) diketahui mengandung flavonoid dan fenolik yang berpotensi sebagai agen antibakteri. Penelitian ini dilakukan secara eksperimental untuk mengetahui kandungan total flavonoid, fenolik, serta aktivitas antibakteri ekstrak etanol daun saga terhadap S. aureus ATCC 25923 dan MRSA ATCC 33591. Ekstraksi dilakukan dengan metode maserasi menggunakan etanol, analisis kandungan senyawa dengan metode kolorimetri AlCl₃ dan Folin-Ciocalteu, serta uji antibakteri dengan metode difusi cakram. Hasil menunjukkan kandungan flavonoid sebesar 13,81 ± 0,71 mg QE/g dan fenolik 42,50 ± 0,35 mg GAE/g, dengan daya hambat masing-masing 4 mm terhadap S. aureusdan 4,25 mm terhadap MRSA. Disimpulkan bahwa meskipun mengandung senyawa aktif, aktivitas antibakteri ekstrak tergolong lemah, kemungkinan akibat keterbatasan difusi senyawa atau keberadaan biofilm.

Staphylococcus aureus infection, especially MRSA strains, is a serious problem due to its resistance to various antibiotics. Saga leaves (Abrus precatorius) are known to contain flavonoids and phenolics that have the potential as antibacterial agents. This study was conducted experimentally to determine the total flavonoid, phenolic content, and antibacterial activity of ethanol extract of saga leaves against S. aureus ATCC 25923 and MRSA ATCC 33591. Extraction was carried out by maceration method using ethanol, analysis of compound content using AlCl₃ and Folin-Ciocalteu colorimetric methods, and antibacterial test using disc diffusion method. The results showed flavonoid content of 13.81 ± 0.71 mg QE/g and phenolic 42.50 ± 0.35 mg GAE/g, with inhibitory power of 4 mm against S. aureusand 4.25 mm against MRSA, respectively. It was concluded that despite containing active compounds, the antibacterial activity of the extract was relatively weak, possibly due to limited diffusion of the compounds or the presence of biofilm.
