PENGARUH PERBEDAAN KONSENTRASI PERCOLL DAN LAMA INKUBASI TERHADAP MOTILITAS DAN ABNORMALITAS SPERMATOZOA X HASIL SEPARASI SEKS PADA KAMBING SAANEN

Penelitian ini bertujuan untuk mengetahui interaksi penggunakan percoll sebagai medium separasi seks dan lama inkubasi terhadap motilitas dan abnormalitas spermatozoa X hasil separasi seks pada kambing Saanen. Materi yang digunakan dalam penelitian yaitu pejantan kambing Saanen berumur 2 tahun dengan bobot 50 Kg. Bahan-bahan yang digunakan berupa semen dari Kambing Saanen yang dikoleksi, medium percoll konsentrasi 35% dan 70% serta 45% dan 90%, Phospat Buffer Saline (PBS) sebagai pengencer medium percoll, aquabidest, tris amino methane sebagai pengencer semen, laktosa, antibiotik penisilin dan streptomycin, raffinose pentatydrate, asam sitrat, kuning telur dan eosin-negrosin. Alat yang digunakan yaitu vagina buatan, micropipet, mikroskop, microtip, object glass, cover glass, sentrifugator, tabung sentrifuse, thermometer, waterbath, erlenmeyer, stopwatch, dan gelas beker. Rancangan penelitian yang digunakan dalam penelitian eksperimental tersebut yaitu Rancangan Acak Kelompok (RAK) pola faktorial 2 x 3 dengan blok yaitu frekuensi penyadapan sebanyak 3 kali yang terdiri dari dua faktor yaitu (g1:35%:70% dan g2:45%:90%) dan faktor lama inkubasi yang terdiri dari tiga level (30,45, dan 60 menit). Data penelitian yang telah diperoleh dianalisis variansi dan dengan uji lanjut Beda Nyata Jujur (BNJ). Hasil penelitian menunjukkan bahwa tidak ada interaksi perbedaan antara konsentrasi percoll dan lama inkubasi terhadap motilitas dan abnormalitas (P>0.05), namun pada faktor lama inkubasi memberikan pengaruhnya masing-masing. Hasil motilitas berdasarkan waktu lama inkubasi diperoleh nilai motilitas 65.50 ± 2.00% - 74.17 ± 2.50%. Berdasarkan waktu lama inkubasi pada abnormalitas diperoleh nilai abnormalitas 10.33 ± 1.50%-13.17 ± 2.00%. Kesimpulan dari penelitian ini adalah motilitas spermatozoa X hasil separasi seks kambing Saanen terbaik pada waktu konsentrasi 35%:70% dengan lama inkubasi 30 menit.

This study aims to determine the effect of the use of percoll as a medium of sex separation and the duration of incubation on the motility and abnormality of spermatozoa X resulting from sex separation in Saanen goats. The material used in the study was a 2-year old Saanen goat male with a weight of 50 Kg. The materials used are cement from Saanen goats that are collected, percoll medium with concentrations of 35% and 70% and 45% and 90%, Phosphate Buffer Saline (PBS) as a diluent of percoll medium, aquabidest, tris amino methane as a diluent of cement, lactose, antibiotics penicillin and streptomycin, raffinose pentatydrate, citric acid, egg yolk and eosin-negrosin. The tools used are artificial vaginas, micropipettes, microscopes, microtips, object glasses, cover glass, centrifuges, centrifuge tubes, thermometers, waterbaths, erlenmeyers, stopwatches, and beakers. The research design used in the experimental study is a Group Random Design (RAK) with a 2 x 3 factorial pattern with blocks, namely the frequency of tapping 3 times consisting of two factors, namely (g1:35%:70% and g2:45%:90%) and the incubation time factor consisting of three levels (30, 45, and 60 minutes). The research data that has been obtained is analyzed by variance and by further testing of Beda Nyata Jujur (BNJ). The results showed that there was no differential interaction between percoll concentration and incubation time on motility and abnormality (P>0.05), but the incubation time factor had its respective effects. The motility results based on the incubation time were obtained with a motility value of 65.50 ± 2.00% - 74.17 ± 2.50%. Based on the long incubation time on the abnormality, the abnormality value was obtained 10.33 ± 1.50%-13.17 ± 2.00%. The conclusion of this study is that the motility and abnormalities of spermatozoa X resulting from sex separation of Saanen goats is best at a concentration time of 35%:70% with an incubation time of 30 minutes.
