PENGARUH STARVASI TERHADAP KEMAMPUAN MIGRASI SEL PUNCA MESENKIMAL TALI PUSAT MANUSIA DENGAN METODE SCRATCH ASSAY

Latar Belakang: Terapi sel punca mesenkimal (SPM) tali pusat manusia tengah berkembang pesat. Namun, masih terdapat tantangan terapi seperti proliferasi dan migrasi yang tidak terkendali. Sehingga, media terkondisi SPM sebagai upaya untuk meminimalisir tantangan tersebut melalui sinkronisasi siklus sel sebelum transplantasi. Tujuan: Penelitian ini bertujuan untuk mengetahui pengaruh starvasi terhadap kemampuan SPM tali pusat manusia terhadap kemampuan migrasi dengan metode scratch assay. Metodologi: penelitian eksperimental dengan pendekatan “post test only with control group” dengan sampel berupa gambar area migrasi dari mikroskop inversi sebanyak 4 lapang pandang disetiap kelompok perlakuan, yaitu kelompok media komplit pada DMEM+FBS (MK), starvasi serum pada media DMEM only (BASAL) dan starvasi asam amino pada media HBSS (HBSS). Serta pengambilan gambar menurut durasi starvasi. Perlakuan diulang sebanyak 1 kali. Kuantifikasi luas penutupan migrasi SPM menggunakan software ImageJ. Analisis data menggunakan One-Way ANOVA. Hasil: Terdapat perbedaan yang signifikan (p<0,05) kemampuan migrasi SPM didalam 3 kelompok pada jam ke-4 dan 8. Terdapat perbedaan yang signifikan (p<0,05) rerata kemampuan migrasi SPM pada media BASAL dan HBSS disetiap periode durasi starvasi. Kesimpulan: Kemampuan migrasi pada kelompok HBSS lebih lambat dibandingkan kelompok lainnya. Durasi starvasi terbaik pada tiga kelompok media pada jam ke-4 dan 8. Background: Human umbilical mesenchymal stem cell (hUMSC) therapy is rapidly advancing. However, challenges persist in therapeutic applications, such as uncontrolled proliferation and migration. Thus, conditioned media for stem cells has been explored as a means to minimize these challenges by synchronizing the cell cycle prior to transplantation. Objective: This study aims to investigate the effect of starvation on the migration ability of human umbilical mesenchymal stem cells (hUMSCs) using the scratch assay method. Methodology: An experimental study with a "post-test only with control group" approach was conducted, with samples consisting of migration area images taken from an inverted microscope at four different fields of view for each treatment group, namely the complete media group in DMEM+FBS (MK), serum starvation in DMEM only (BASAL), andasam amino starvation in HBSS media (HBSS). Images were captured according to the starvation duration, and treatments were repeated once. Migration area quantification was performed using ImageJ software. Data analysis was conducted using One-Way ANOVA. Results: A significant difference (p<0.05) in migration ability was observed in the three groups at the 4th and 8th hours. A significant difference (p<0.05) in the average migration ability of hUMSCs was noted in the BASAL and HBSS media across different starvation durations. Conclusion: The migration ability in the HBSS group was slower compared to the other groups. The optimal starvation duration across all media groups was observed at the 4th and 8th hours.
