PENGARUH BERBAGAI MODEL SLEEP DEPRIVATION METODE SINGLE PLATFORM TERHADAP KADAR MALONDIALDEHID (MDA) TIKUS PUTIH (Rattus norvegicus) JANTAN Faradina Gita Paramasari1, Fitranto Arjadi 2, Nur Signa Aini Gumilas 3, Ika Murti Harini 4, Yudhi Wibowo5 Fakultas Kedokteran, Universitas Jenderal Soedirman, Purwokerta Indonesia Email: faradinajpr@gmail.com ABSTRAK Latar belakang: Sleep deprivation (kurang tidur) dapat menyebabkan stres oksidatif, meningkatkan peroksidasi lipid, dan kadar malondialdehid (MDA) yang menjadi penanda stres oksidatif. Penelitian ini menggunakan metode single platform untuk mengevaluasi efek sleep deprivation pada tikus putih jantan. Tujuan: Mengetahui pengaruh berbagai model sleep deprivation terhadap kadar MDA pada tikus putih (Rattus norvegicus). Metodologi: Penelitian ini adalah eksperimen dengan desain posttest-only with control group. Sebanyak 30 tikus dibagi menjadi lima kelompok: kontrol, PSD (paradoxical sleep deprivation), TSD (total sleep deprivation), PSD+SR (dengan sleep recovery), dan TSD+SR. Sleep deprivation dilakukan selama lima hari, dan kadar MDA diukur menggunakan metode ELISA. Analisis data menggunakan One Way ANOVA dan uji post hoc untuk membandingkan antar kelompok. Hasil: Terdapat perbedaan kadar MDA yang signifikan antar kelompok (p=0,045). Kelompok PSD+SR memiliki kadar MDA tertinggi (1,969 ± 0,79 nmol/mL), diikuti oleh PSD, TSD+SR, TSD, dan kontrol. Analisis post hoc menunjukkan kelompok PSD+SR berbeda signifikan dengan kontrol dan TSD. Kesimpulan: Model sleep deprivation memengaruhi kadar MDA secara signifikan, dengan model PSD+SR menghasilkan kadar MDA tertinggi. This study applied the single platform method to assess the effects of sleep deprivation in male white rats. Objective: To evaluate the effects of various sleep deprivation models on MDA levels in male white rats (Rattus norvegicus). Methodology: This experimental study employed a posttest-only with control group design. Thirty rats were divided into five groups: control, PSD (paradoxical sleep deprivation), TSD (total sleep deprivation), PSD+SR (with sleep recovery), and TSD+SR. Sleep deprivation was performed for five days, and MDA levels were measured using the ELISA method. Data were analyzed using One Way ANOVA and post hoc tests for intergroup comparisons. Results: There were significant differences in MDA levels among the groups (p=0.045). The PSD+SR group exhibited the highest MDA levels (1.969 ± 0.79 nmol/mL), followed by PSD, TSD+SR, TSD, and control groups. Post hoc analysis revealed significant differences between PSD+SR, control, and TSD groups. Conclusion: Sleep deprivation models significantly affect MDA levels, with the PSD+SR model producing the highest MDA levels.
