PENGARUH SENTRIFUGASI GRADIEN DENSITAS PERCOLL DAN LAMA INKUBASI TERHADAP MORFOMETRI DAN MEMBRAN PLASMA UTUH HASIL SEXING SPERMATOZOA KAMBING PERANAKAN ETAWAH

Penelitian ini berjudul Pengaruh Sentrifugasi Gradien Densitas Percoll dan Lama Inkubasi Terhadap Morfometri dan Membran Plasma Utuh Hasil Sexing Spermatozoa Kambing Peranakan Etawah. Tujuan penelitian dilakukan untuk mengkaji interaksi gradien percoll dan lama inkubasi terhadap morfometri dan Membran Plasma Utuh (MPU) hasil separasi seks kambing Peranakan Etawah (PE). Materi yang digunakan untuk penelitian yaitu semen kambing Peranakan Etawah (PE) jantan sebanyak 2 ekor dengan umur 3-4 tahun. Rancangan penelitian dilakukan menggunakan Rancangan Acak Kelompok (RAK) pola faktorial 2 x 3 dengan ulangan frekuensi penyadapan sebanyak 3 kali. Hasil analisis variansi perlakuan yang berpengaruh nyata kemudian dilakukan uji lanjut dengan uji Duncan’s Multiple Range Test (DMRT). Lokasi penelitian berada di Laboratorium Kesehatan Ternak dan Experimental Farm Fakultas Peternakan, Universitas Jenderal Soedirman, Purwokerto, Kabupaten Banyumas, Jawa Tengah. Hasil pengukuran morfometri rata-rata spermatozoa unsexing 30.88 µm2 , spermatozoa sexing fraksi atas dan fraksi bawah masing- masing 28.44 µm2 dan 32.63 µm2 . Hasil dari penelitian keutuhan membran plasma spermatozoa X diperoleh rataan sebesar X1 = 66.61% dan X2 = 68.57% sedangkan spermatozoa Y diperoleh rataan sebesar Y1 = 64,87% dan Y2 = 66.96%. Hasil perhitungan statistic dengan uji t-student menunjukkan bahwa spermatozoa fraksi bawah (X) memberikan perbedaan nyata (P<0,05) lebih besar dibanding spermatozoa fraksi atas (Y). Berdasaran analisis variansi diperoleh hasil tidak ada interaksi antara faktor gradien dan lama inkubasi (P>0,05) terhadap morfometri dan Membran Plasma Utuh (MPU). Faktor gradien berpengaruh nyata (P<0,05) terhadap morfometri fraksi atas (Y) dan bawah (X), sedangkan faktor gradien dan lama inkubasi masing-masing berpengaruh nyata (P<0,05) terhadap Membran Plasma Utuh (MPU) fraksi atas (Y) dan bawah (X). Kesimpulan yang didapat dari penelitian ini yaitu separasi seks menggunakan medium gradien percoll 45:60% dengan lama inkubasi 30 menit mampu memisah dengan baik morfometrik spermatozoa fraksi atas (Y) maupun fraksi bawah (X), bahkan diperoleh persentase membran plasma utuh (MPU) tertinggi baik untuk fraksi atas dan bawah.

This research is entitled The Effect of Percoll Density Gradient Centrifugation (SGDP) and Incubation Length on Morphometry and Intact Plasma Membrane (MPU) Results from Sexing Spermatozoa of Etawah Crossbreed Goats. The aim of the research was to examine the interaction of Percoll gradient and incubation time on the morphometry and Intact Plasma Membrane (MPU) results of sex separation of Etawah Peranakan (PE) goats. The material used for the research was the semen of 2 male Peranakan Etawah (PE) goats aged 3-4 years. The research design was carried out using a Randomized Block Design (RAK) with a 2 x 3 factorial pattern with 3 repetitions of the tapping frequency. The results of the variance analysis of treatments that had a significant effect were then further tested using Duncan's Multiple Range Test (DMRT). The research location is at the Animal Health and Experimental Farm Laboratory, Faculty of Animal Husbandry, Jenderal Soedirman University, Purwokerto, Banyumas Regency, Central Java. The average morphometric measurement results for unsexed spermatozoa were 30.88 µm2 , sexed spermatozoa in the upper and lower fractions were 28.44 µm2 and 32.63 µm2 , respectively. The results of research on the integrity of the plasma membrane of X spermatozoa obtained an average of X1 = 66.61% and X2 = 68.57%, whereas integrity plasma membrane Y of Y1 = 64,87% and Y2 = 66.96%. The results of statistical calculations using the Student's t-test showed that the lower fraction spermatozoa (X) gave a significantly greater difference (P<0.05) than the upper fraction spermatozoa (Y). Based on analysis of variance, the results showed that there was no interaction between gradient factors and incubation time (P>0.05) on morphometry and Intact Plasma Membrane (MPU). The gradient factor had a significant effect (P<0.05) on the morphometry of the top (Y) and bottom (X) fractions, while the gradient factor and incubation time each had a significant effect (P<0.05) on the Intact Plasma Membrane (MPU) fraction top (Y) and bottom (X). The conclusion obtained from this research is that sex separation using a 45:60% Percoll gradient medium with an incubation time of 30 minutes was able to separate both upper (Y) and lower (X) fractions of spermatozoa morphometrically, and even obtained the highest percentage of intact plasma membrane (MPU), both for the upper and lower fractions.
