PENGARUH PENGGUNAAN GRADIEN BOVINE SERUM ALBUMIN SEBAGAI MEDIUM SEPARASI SEKS DAN LAMA INKUBASI TERHADAP MOTILITAS DAN ABNORMALITAS SPERMATOZOA KAMBING PERANAKAN ETAWAH

Penelitian bertujuan untuk mengetahui pengaruh penggunaan gradien bovine serum albumin sebagai medium separasi seks dan lama inkubasi terhadap motilitas dan abnormalitas spermatozoa kambing Peranakan Etawah. Materi penelitian yang digunakan yaitu 2 ekor pejantan kambing Peranakan Etawah, medium Bovine Serum Albumin (BSA), NaCl fisiologis, dan eosin-negrosin. Pakan tambahan yang digunakan yaitu tauge 250 gram/hari. Metode penelitian yang digunakan yaitu Rancangan Acak Kelompok (RAK) pola faktorial 2 x 2 x 3 dengan ulangan sebanyak 3 kali dan menggunakan 3 faktor yaitu fraksi (F) terdiri dari dua level (atas dan bawah), faktor gradien (G) terdiri dari dua level (5:15% dan 10:20%) serta faktor lama inkubasi (T) terdiri dari tiga level (30, 45 dan 60 menit) sehingga di dapatkan 12 kombinasi perlakuan. Variabel yang diukur adalah motilitas dan abnormalitas spermatozoa setelah separasi seks. Data penelitian yang telah diperoleh dianalisis variansi dan menggunakan uji lanjut Duncan’s Multiple Range Test (DMRT). Hasil penelitian menunjukkan bahwa tidak ada interaksi antara fraksi atas dan bawah, medium BSA dan perbedaan lama inkubasi terhadap motilitas dan abnormalitas (P > 0,05), namun setiap faktor memberikan pengaruhnya masing-masing. Berdasarkan faktor fraksi nilai motilitas dan abnormalitas masing-masing yaitu 66,05 – 70,13% dan 10,25 – 10,29%. Berdasarkan faktor gradien BSA didapatkan nilai motilitas dan abnormalitas sebesar 66,01 – 70,18% dan 10,20 – 11,34%. Nilai motilitas dan abnormalitas berdasarkan faktor lama inkubasi yaitu 64,25 – 71,45% dan 10,11 – 11,37%. Disimpulkan bahwa motilitas spermatozoa kambing Peranakan Etawah hasil separasi seks tertinggi pada fraksi atas (Spermatozoa X) perbandingan gradien 5:15 dengan lama inkubasi 45 menit yaitu 74,59 ± 1,52% dan abnormalitas spermatozoa kambing Peranakan Etawah hasil separasi seks terendah pada fraksi bawah (Spermatozoa Y) perbandingan gradien 10:20 dengan lama inkubasi 45 menit pada suhu 37°C yaitu 8,80 ± 0,96%.

This study aims to determine the effect of using bovine serum albumin gradient as a sex separation medium and incubation time on the motility and abnormality of Etawah crossbred goat spermatozoa. The research materials used were 2 male Etawah crossbreed goats, Bovine Serum Albumin (BSA) medium, physiological NaCl, and eosin-nigrosin. The additional feed used was bean sprouts 250 g/day. The research method used was a Randomized Block Design (RBD) factorial pattern 2 x 2 x 3 with 3 replications and using 3 factors, namely fraction (F) consisting of two levels (top and bottom), gradient factor (G) consisting of two levels (5:15% and 10:20%) and incubation time factor (T) consisting of three levels (30, 45 and 60 minutes) so that 12 treatment combinations were obtained. The variables measured were motility and spermatozoa abnormality after sex separation. The research data obtained were analyzed for variance and using the Duncan's Multiple Range Test (DMRT) further test. The results showed that there was no interaction between the upper and lower fractions, BSA medium and differences in incubation time on motility and abnormality (P> 0.05), but each factor had its own influence. Based on the fraction factor, the motility and abnormality values were 66.05 - 70.13% and 10.25 - 10.29%, respectively. Based on the BSA gradient factor, the motility and abnormality values were 66.01 - 70.18% and 10.20 - 11.34%. The motility and abnormality values based on the incubation time factor were 64.25 - 71.45% and 10.11 - 11.37%. The conclusion obtained from this study is that the highest motility of Etawah crossbred goat spermatozoa resulting from sex separation was in the upper fraction (Spermatozoa X) with a gradient ratio of 5:15 with an incubation period of 45 minutes, namely 74.59 ± 1.52% and the lowest abnormality of Etawah crossbred goat spermatozoa resulting from sex separation was in the lower fraction (Spermatozoa Y) with a gradient ratio of 10:20 with an incubation period of 45 minutes at a temperature of 37°C, namely 8.80 ± 0.96%.
