INDUKSI MUTASI DENGAN KOLKISIN PADA KENTANG (Solanum tuberosum L.) VARIETAS TEDJO MZ SECARA IN VITRO

Penelitian ini bertujuan untuk mengetahui induksi mutasi dengan kolkisin pada kentang (Solanum tuberosum L.) varietas Tedjo MZ secara in vitro. Percobaan yang digunakan yaitu menggunakan metode rancangan acak kelompok (RAK) non faktorial yang terdiri atas 1 perlakuan yakni konsentrasi kolkisin (K) dengan 4 taraf yaitu K0 (0 ppm), K1 (20 ppm), K2 (40 ppm), dan K3 (60 ppm) serta setiap taraf perlakuan diulang 3 kali. Satu unit ulangan terdiri dari 5 botol kultur dengan 2 planlet. Variabel yang diamati yaitu persentase hidup planlet normal, tinggi planlet, bobot planlet, waktu muncul tunas, jumlah tunas aksilar, waktu muncul akar, jumlah akar, panjang akar, jumlah buku, jumlah daun, luas daun, deskripsi warna daun, dan deskripsi warna batang. Data yang diperoleh dianalisis menggunakan oneway ANOVA dan apabila berbeda nyata dilakukan uji Least Significant Difference (LSD) pada taraf 5%. Berdasarkan hasil penelitian tentang induksi mutasi dengan kolkisin pada kentang varietas Tedjo MZ secara in vitro, didapatkan bahwa: 1) Konsentrasi kolkisin berpengaruh terhadap pertumbuhan dan perkembangan planlet kentang varietas Tedjo MZ secara in vitro. Konsentrasi kolkisin yang diberikan mempercepat waktu muncul akar, menurunkan jumlah hidup planlet normal, memberikan pengaruh dengan jumlah yang beragam pada jumlah tunas aksilar, jumlah akar, jumlah buku, jumlah daun, warna daun, dan warna batang, 2) Konsentrasi kolkisin 20 ppm memberikan pengaruh yang baik terhadap persentase planlet hidup planlet normal, jumlah tunas aksilar, jumlah akar, jumlah buku, jumlah daun, warna daun, dan warna batang, sedangkan konsentrasi kolkisin 40 ppm memberikan pengaruh yang sangat baik terhadap waktu muncul akar.

This research aims to determine the induction of mutations with colchicine in potatoes (Solanum tuberosum L.) of the Tedjo MZ variety in vitro. The experiment used was a non-factorial randomized block design (RAK) method consisting of 1 treatment, namely colchicine (K) concentration with four levels, namely K0 (0 ppm), K1 (20 ppm), K2 (40 ppm), and K3 ( 60 ppm) and each treatment level was repeated three times. One replication unit consists of 5 culture bottles with two plantlets. The variables observed were the percentage of normal plantlet life, plantlet height, plantlet weight, shoot emergence time, number of axillary shoots, root emergence time, number of roots, root length, number of nodes, number of leaves, leaf area, description of leaf color, and description of stem color. The data obtained were analyzed using one-way ANOVA, and if they were significantly different, a Least Significant Difference (LSD) test was carried out at the 5% level. Based on the results of research on mutation induction with colchicine in the Tedjo MZ potato variety in vitro, it was found that: 1) Colchicine concentration affected the growth and development of plantlets of the Tedjo MZ potato variety in vitro. The concentration of colchicine given accelerates the time for roots to emerge, reduces the number of normal plantlet life, and has varying amounts of influence on the number of axillary shoots, number of roots, number of nodes, number of leaves, leaf color, and stem color, 2) Colchicine concentration of 20 ppm has an effect which was good for the percentage of live plantlets of normal plantlets, number of axillary shoots, number of roots, number of nodes, number of leaves, leaf color and stem color, while the colchicine concentration of 40 ppm had an excellent influence on the time of root emergence.
