EKSPLORASI DAN UJI POTENSI BAKTERI REMEDIATOR PESTISIDA SINTETIK GLIFOSAT DARI PERAKARAN TANAMAN PADI (Oryza sativa) DI KABUPATEN PEMALANG

Isolat bakteri yang digunakan dalam penelitian ini berasal dari lahan sawah di Desa Mandiraja, Kecamatan Moga dan Kabupaten Pemalang. Isolat yang didapatkan diberi kode isolat KT 1 (a), KT 2 (a), KM (a), KB 1 (a), dan KB 2 (a). Penelitian dilakukan beberapa tahap yaitu pengambilan sampel, pengukuran pH, isolasi bakteri, pengamatan morfologi koloni, pewarnaan gram dan spora, uji resistensi, uji kurva pertumbuhan, dan uji remediasi. Hasil isolasi bakteri dengan medium NA yang ditambah glifosat 1 ppm diperoleh lima isolat bakteri yaitu KT I (a), KT 2 (a), KM (a), KB 1 (a), dan KB 2 (a). Berdasarkan uji TPC (Total Plate Count) dengan rentang jumlah koloni 25-250 diperoleh jumlah koloni bakteri berkisar 22,5 x 105 – 385,35 x 105 CFU/ml. Karakteristik morfologi koloni dari kelima isolat bakteri didominasi dengan ukuran kecil, bentuk bulat, tekstur halus, warna putih, tepi halus, dan elevasi datar. Berdasarkan pewarnaan gram, semua isolat merupakan gram negatif. Semua isolat memiliki kemampuan untuk membentuk spora. Pada uji kurva pertumbuhan tanpa cemaran glifosat didapatkan isolat KB 2 (a) memiliki waktu generasi isolat paling baik yaitu 1 jam 24 menit. Hasil uji resistensi glifosat dan uji pertumbuhan bakteri tercekam glifosat paling baik yaitu isolat KM (a) pada konsentrasi 2 ppm. Uji remediasi menunjukkan bahwa KB 2 (a) merupakan isolat yang mempunyai kemampuan paling besar dalam mengurangi kadar glifosat.

The bacterial isolates used in this study came from paddy fields in Mandiraja Village, Moga District and Pemalang Regency. The isolates obtained were coded as KT 1 (a), KT 2 (a), KM (a), KB 1 (a), and KB 2 (a). The research was carried out in several stages, namely sampling, measuring pH, isolating bacteria, observing colony morphology, gram and spore staining, resistance testing, growth curve testing, and remediation testing. The results of bacterial isolation with NA medium added with 1 ppm glyphosate obtained five bacterial isolates namely KT I (a), KT 2 (a), KM (a), KB 1 (a), and KB 2 (a). Based on the TPC (Total Plate Count) test with a range of 25-250 colonies, the number of bacterial colonies ranged from 22.5 x 105 – 385.35 x 105 CFU/ml. Colony morphological characteristics of the five bacterial isolates were dominated by small size, round shape, smooth texture, white color, smooth edges, and flat elevation. Based on gram staining, all isolates were gram negative. All isolates have the ability to form spores. In the growth curve test without glyphosate contamination, it was found that KB 2 (a) isolates had the best isolate generation time of 1 hour 24 minutes. The best results of the glyphosate resistance test and the best glyphosate-stressed bacterial growth test were isolate KM (a) at a concentration of 2 ppm. The remediation test showed that KB 2 (a) was the isolate that had the greatest ability to reduce glyphosate levels.
