PENGARUH PENGGUNAAN JENIS PENGENCER SEMEN YANG BERBEDA TERHADAP MOTILITAS DAN FERTILITAS SPERMATOZOA AYAM KAMPUNG

Penelitian ini dilaksanakan di Teaching and Experimental Farm, Fakultas Peternakan, Universitas Jenderal Soedirman dari tanggal 2 Oktober sampai dengan 20 Oktober 2022. Tujuan penelitian ini adalah untuk mengetahui pengaruh penggunaan jenis pengencer semen terhadap motilitas dan fertilitas spermatozoa ayam kampung. Materi dan Metode. Materi yang digunakan pada penelitian adalah semen segar yang berasal dari 12 ekor ayam kampung jantan berumur 1 - 1,5 tahun, 50 ekor ayam niaga petelur umur 90 minggu, susu skim, NaCl fisiologis, ringer laktat, dan glukosa. Penelitian ini menggunakan 2 rancangan penelitian, Rancangan Acak Kelompok (RAK) untuk motilitas dengan 5 jenis pengencer sebagai perlakuan dan 5 periode penyadapan sebagai blok, dan Rancangan Acak Lengkap (RAL) untuk fertilitas dengan 5 jenis pengencer sebagai perlakuan dan terdapat 5 kali ulangan. Perlakuan yang diberikan adalah P0 = susu skim 100%, P1 = susu skim 80% + NaCl fisiologis 20%, P2 = susu skim 80% + ringer laktat 20%, P3 = susu skim 80% + NaCl fisiologis 20% + glukosa 50 mM, P4 = susu skim 80% + ringer laktat 20% + glukosa 50 mM. Data dianalisis menggunakan analisis variansi kemudian jika terdapat pengaruh atau perbedaan yang signifikan maka dilakukan uji lanjut Beda Nyata Jujur (BNJ). Hasil. Hasil ANAVA menunjukan penggunaan jenis pengencer semen yang berbeda berpengaruh nyata terhadap motilitas spermatozoa (P<0.05) sedangkan pada fertilitas berpengaruh tidak nyata (P>0.05). Hasil penelitian menunjukan bahwa rata-rata persentase nilai motilitas spermatozoa ayam kampung berkisar antara 83.60-90.40% (nilai tertinggi pada perlakuan P3 susu skim 80% + NaCl fisiologis 20% + glukosa 50 mM dengan nilai 90.40% dan terendah pada perlakuan P0 susu skim 100% dengan nilai 83.60%). Rata-rata presentase nilai fertilitas spermatozoa ayam kampung berkisar antara 62.29-80.79% (nilai tertinggi pada perlakuan P0 susu skim 100% dengan nilai 80.79% dan terendah pada perlakuan P1 susu skim 80% + NaCl fisiologis 20% dengan nilai 62.29%). Kesimpulan. motilitas tertinggi pada pengencer susu skim 80% + NaCl 20% + glukosa 20 mM, dan fertilitas terbaik pada pengencer susu skim 100%.

The research was conducted at the Experimental and Teaching Farm, Faculty of Animal Science, Jenderal Soedirman University from 2 October until 20 October 2022. The purpose of this study was to determine the effect of using a type of semen diluent on the motility and fertility kampung rooster spermatozoa. Material and Methods. The material used in this research include fresh semen from 12 kampung rooster age ranging from 1-1,5 years old, 50 commercial strains of egg-laying Isa Brown hens age 90 weeks, skim milk, physiological saline, ringer’s lactate, and glucose. Randomized Block Design (RBD) for motility with 5 types of diluent as treatment and 5 collection periods as a block, and Completely Randomized Design (CRD) for fertility with 5 types of diluent as treatment and there were 5 replications. The treatments given were P0 = skim milk 100%, P1 = skim milk 80% + physiological saline 20%, P2 = skim milk 80% + ringer’s lactate 20%, P3 = skim milk 80% + physiological saline 20% + glucose 50 mM, P4 = skim milk 80% + ringer’s lactate 20% + glucose 50 mM. The data were analyzed using analysis of variance then if there was a significant influence or difference then a follow up test was carried out for honest significant difference (HSD). Result. ANOVA results showed that the use of different types of semen diluent had a significant effect on spermatozoa motility (P<0.05), while fertility had no significant effect (P>0.05). Result showed that the average percentage value of kampung rooster spermatozoa motility ranged from 83.60-90.40% (the highest value was in P3 treatment of skim milk 80% + physiological saline 20% + glucose 50 mM with a value of 90.40% and the lowest was in P0 treatment of skim milk 100% with a value of 83.60%). While the average percentage value of kampung rooster spermatozoa fertility was 62.29-80.79% (the highest value was in P0 treatment of skim milk 100% with a value of 80.79% and the lowest was in P1 treatment of skim milk 80% + physiological saline 20% with a value of 62.29%). Conclusion. The highest motility was in the diluent of skim milk 80% + physiological saline 20% + glucose 50 mM, and the best fertility was in the diluent of skim milk 100%.
