PENGARUH PENGGUNAAN PENGENCER SEMEN DENGAN BAHAN DASAR SUSU SKIM TERHADAP MOTILITAS DAN FERTILITAS SPERMATOZOA AYAM KAMPUNG

Penelitian ini bertujuan untuk mengetahui pengaruh penggunaan pengencer semen dengan bahan dasar susu skim terhadap motilitas dan fertilitas spermatozoa ayam kampung. Materi yang digunakan pada penelitian ini antara lain semen segar dari 10 ekor ayam kampung jantan berumur 1,5 - 2 tahun, 40 ekor ayam niaga petelur berumur 71 minggu, susu skim bubuk, aquadest, aquabidest, NaCl fisiologis, dan ringer laktat. Penelitian ini terdiri dari dua rancangan percobaan yaitu Rancangan Acak Kelompok (RAK) untuk motilitas dengan 4 perlakuan dan 6 kali ulangan dan Rancangan Acak Lengkap (RAL) untuk fertilitas dengan 4 perlakuan dan 5 kali ulangan. Perlakuan yang diberikan adalah P1 = susu skim + pelarut aquadest, P2 = susu skim + pelarut aquabidest, P3 = susu skim (perlakuan P2) 80% + NaCl fisiologis 20%, P4 = susu skim (perlakuan P2) 80% + ringer laktat 20%. Data dianalisis menggunakan analisis variansi dan uji lanjut Beda Nyata Jujur (BNJ). Hasil penelitian menunjukkan bahwa rata-rata persentase nilai motilitas spermatozoa ayam kampung berkisar antara 84,67-90,00% (nilai tertinggi pada perlakuan P4 dan terendah pada perlakuan P3), sedangkan rata-rata persentase nilai fertilitas spermatozoa ayam kampung berkisar antara 88,45-95,56% (nilai tertinggi pada perlakuan P4 dan terendah pada perlakuan P1). Dapat disimpulkan bahwa pengunaan pengencer semen susu skim (perlakuan P2) 80% + ringer laktat 20% memberikan hasil yang tertinggi dalam mempertahankan motilitas dan fertilitas spermatozoa ayam kampung.

The purpose of this study was to determine the motility and fertility of Kampung rooster spermatozoa diluted in several skimmed-milk based semen diluents. The materials used in this research include fresh semen from 10 kampung rooster age ranging from 1.5-2 years old, 40 commercial strains of egg-laying Isa Brown hens age 71 weeks, skim milk, aquadest, aquabidest, physiological saline, and ringer’s lactate. This study consisted of 2 experimental designs, a randomized block design (RBD) for motility with 4 types of treatments and 6 replications and complete randomized design (CRD) for fertility with 4 types of treatments and 5 replications. The treatments given were P1 = skim milk + aquadest solvent, P2 = skim milk + aquabidest solvent, P3 = skim milk (P2 treatment) 80% + 20% physiological saline, P4 = skim milk (P2 treatment) 80% + 20% ringer’s lactate. The data were analyzed using analysis of variance and advanced test of honestly significant difference (HSD). Result showed that the average percentage value of kampung rooster spermatozoa motility ranged from 84.67-90.00% (the highest value was in treatment P4 and the lowest was in treatment P3), while the average percentage value of kampung rooster spermatozoa fertility was 88.54-95.56% (the higest value was in treatment P4 and the lowest was in treatment P1). It can be concluded that the use of skim milk (treatment P2) 80% + 20% ringer's lactate gave the highest results in maintaining the motility and fertility of kampung rooster spermatozoa.
