FORMULASI PASTA GIGI EKSTRAK ETANOL DAUN SIRSAK (Annona muricata L.) DENGAN VARIASI KONSENTRASI Na CMC SEBAGAI ANTIBAKTERI Streptococcus mutans

Latar Belakang : Karies gigi umumnya disebabkan oleh kebersihan mulut yang buruk sehingga terjadi akumulasi plak yang mengandung berbagai macam bakteri, bakteri yang berperan penting dalam penyakit ini adalah bakteri Streptococcus mutans. Daun sirsak terbukti memiliki daya antibakteri pada S. mutans. Pasta gigi merupakan sediaan yang dapat digunakan untuk membersihkan gigi dari plak. Viskositas merupakan parameter penting dalam pembuatan pasta gigi. Tujuan penelitian ini untuk mengetahui pengaruh variasi konsentrasi Na CMC terhadap karakteristik fisikokimia pasta gigi ekstrak etanol daun sirsak (Annona muricata L.) sebagai antibakteri S. mutans. Metodologi : Pasta gigi diformulasikan menjadi tiga formula dengan variasi konsentrasi Na CMC 1%, 1,5%, dan 2%. Dilakukan uji sifat fisikokimia dan stabilitas sediaan yaitu meliputi uji organoleptis, homogenitas, pH, viskositas, daya sebar, pembentukan busa dan uji freeze-thaw. Hasil dianalisis secara deskriptif dan One way ANOVA. Uji aktivitas antibakteri dilakukan dengan metode difusi sumuran. Hasil Penelitian : Peningkatan konsentrasi Na CMC dapat meningkatkan viskositas dan kemampuan pembentukan busa, menurunkan daya sebar, dan tidak mempengaruhi pH sediaan. Sediaan pasta gigi yang dihasilkan stabil secara fisik. Pengujian antibakteri menunjukkan hasil formula 1 memiliki daya hambat bakteri sebesar 9,74 mm, sedangkan formula 2 dan formula 3 tidak menghasilkan daya hambat. Kontrol negatif dan kontrol positif menghasilkan daya hambat bakteri yang kuat. Kesimpulan : Variasi konsentrasi Na CMC pada pasta gigi ekstrak etanol daun sirsak mempengaruhi konsistensi sediaan, viskositas, daya sebar, dan daya busa. Semakin tinggi konsentrasi Na CMC yang digunakan menyebabkan peningkatan viskositas dan pembentukan busa, namun menurunkan daya sebar sediaan. Formula 1 (Na CMC 1%) memiliki aktivitas antibakteri terhadap S.mutans.

Background: Dental caries is generally caused by poor oral hygiene that causes accumulation of plaque which contains various kinds of bacteria, one of the bacteria that plays an important role in this disease is Streptococcus mutans. Soursop leaves have been shown to have antibacterial activity against Streptococcus mutans. Toothpaste is a cosmeceutical that can be used to cleanse teeth from plaque. Viscosity is an important parameter in the manufacture of toothpaste. The purpose of this study is to determine the effect of various concentrations of Na CMC as a gelling agent on the physicochemical characteristics of toothpaste with soursop (Annona muricata L.) leaf ethanol extract as an antibacterial against Streptococcus mutans. Methodology: Toothpaste is formulated into three formulas with variations of Na CMC levels in 1%, 1.5%, and 2%. Furthermore, the physicochemical and stability of the toothpaste were tested, which included organoleptic tests, homogeneity, pH, viscosity, spreadability, foaming power and freeze-thaw tests. Results were analyzed descripetively and using One way ANOVA. All formulas, negative control and positive control were tested for antibacterial activity by the well diffusion method. Research Results: The results showed that increasing the concentration of Na CMC could increase the viscosity and foaming power, but decrease the spreadability, and did not affect the pH of the toothpaste. Result of stability test is the toothpaste were physically stable. Antibacterial test showed that formula 1 had a bacterial inhibitory on Streptococcus mutans about 9.74 mm, while formula 2 and formula 3 did not shown any bacterial inhibition. Negative control and positive control have strong bacterial inhibition. Conclusion: Variations in the concentration of Na CMC as a gelling agent in soursop leaf ethanol extract toothpaste affect the consistency of the toothpaste, viscosity, spreadability, and foaming power. The higher concentration of Na CMC that used, causes an increase in viscosity and foaming power, but decrease the spreadability of the toothpaste. Formula 1 with 1% level of Na CMC concentration has antibacterial activity against Streptococcus mutans.
