PENGARUH SUPLEMENTASI PROBIOTIK TERHADAP JUMLAH LEUKOSIT DAN DIFERENSIAL LEUKOSIT PADA AYAM NIAGA PETELUR

ADITYA ALDU HALIM. Penelitian yang berjudul “Pengaruh suplementasi probiotik terhadap jumlah leukosit dan diferensial leukosit pada ayam niaga petelur”. Tujuan penelitian untuk mengetahui pengaruh suplementasi probiotik terhadap jumlah leukosit dan diferensial leukosit ayam niaga petelur strain Isa brown serta untuk mengetahui kadar suplementasi probiotik yang optimal untuk pemeliharaan ayam niaga petelur. Materi penelitian terdiri dari ayam niaga petelur berumur 54 minggu strain Isa brown sebanyak 80 ekor yang dipelihara selama 8 minggu, kandang batere, komponen peralatan kandang, pakan periode produksi dengan penambahan probiotik, air minum secara ad libitum, bahan dan peralatan analisis kimia darah. Ransum pakan periode produksi terdiri atas jagung giling (50%), dedak (25%) dan konsentrat (25%) dengan kandungan nutrient energy 2900 kcal/kg, protein kasar 17,5%, Ca 3%, P 1,6%. Penelitian dilakukan dengan metode eksperimental yaitu perlakuan suplementasi probiotik. Rancangan penelitian yang digunakan adalah rancangan acak lengkap (RAL), menggunakan uji saphiro-Wilk (uji normalitas), uji homogenitas dengan metode uji Levene dan uji Kruskal-Wallis dengan 4 perlakuan dan 5 kali ulangan. Perlakuan P0: penambahan probiotik pada pakan 0%, P1: penambahan probiotik pada pakan 0,5%, P2: penambahan probiotik pada pakan 1% dan penambahan probiotik pada pakan 1,5%. Hasil penelitian jumlah leukosit P0 (77560 ± 17599,51 /µl ); P1 (76200 ± 28890,65/µl ); P2 (68580 ± 4140,89 /µl ); dan P3 (80980 ± 48301,26 /µl ). Hasil penelitian jumlah leukosit granulosit P0 (5040 ± 4907,443 /µl ), P1 (2200 ± 877,49 /µl ), P2 (2239,8 ± 881,79 /µl) dan P3 (6000 ± 8002,18 /µl). Hasil penelitian jumlah leukosit agranulosit P0 (75940 ± 17956,84 /µl ), P1 (74000 ± 28162,83 /µl), P2 (66340 ± 3744,07 /µl) dan P3 (85980 ± 24346,60 /µl). Hasil analisis variansi menunjukkan bahwa suplementasi probiotik tidak berpengaruh nyata terhadap jumlah leukosit, jumlah granula leukosit dan jumlah agranula leukosit. Kesimpulannya pemberian perlakuan dengan tingkat suplementasi probiotiki sampai dengan tingkat level tertinggi tidak berpengaruh nyata terhadap jumlah leukosit dan jumlah diferensial leukosit. Pemberian suplementasi probiotik yang optimal pada level 1,5%.

ADITYA ALDU HALIM. The study entitled "The effect of probiotic supplementation on the number of leukocytes and differential leukocytes in laying hens". The purpose of this study to determine the effect of probiotic supplementation on the leukocyte count and leukocyte differential of laying hens strain Isa brown and to determine the optimal level of probiotic supplementation for the maintenance of laying hens. The research material consisted of 80 laying hens aged 54 weeks of Isa brown strain that were reared for 8 weeks, battery cages, components of cage equipment, production period feed with the addition of probiotics, drinking water ad libitum, materials and equipment for blood chemistry analysis. The feed ration for the production period consisted of milled corn (50%), bran (25%) and concentrate (25%) with nutrient energy content of 2900 kcal/kg, crude protein 17.5%, Ca 3%, P 1.6%. The study was conducted using an experimental method, namely the treatment of probiotic supplementation. The research design used was a completely randomized design (CRD), using the saphiro-Wilk test (normality test), homogeneity test using the Levene test method and the Kruskal-Wallis test with 4 treatments and 5 replications. Treatment P0: addition of probiotics to feed 0%, P1: addition of probiotics to feed 0.5%, P2: addition of probiotics to feed 1% and addition of probiotics to feed 1.5%. The results of the study were the number of P0 leukocytes (77560 ± 17599.51 /µl); P1 (76200 ± 28890.65/µl ); P2 (68580 ± 4140.89 /µl); and P3 (80980 ± 48301.26 /µl ). The results of the study were the number of granulocyte leukocytes P0 (5040 ± 4907.443 /µl), P1 (2200 ± 877.49 /µl), P2 (2239.8 ± 881.79 /µl) and P3 (6000 ± 8002.18 /µl) . The results showed that the number of agranulocytes was P0 (75940 ± 17956.84 /µl), P1 (74000 ± 28162.83 /µl), P2 (66340 ± 3744.07 /µl) and P3 (85980 ± 24346.60 /µl). The results of the analysis of variance showed that probiotic supplementation had no significant effect on the number of leukocytes, the number of leukocyte granules and the number of leukocyte agranules. In conclusion, treatment with probiotic supplementation levels up to the highest level did not significantly affect the number of leukocytes and the number of differential leukocytes. Optimal probiotic supplementation at the level of 1.5%.
