AKTIVITAS TRIPSIN-LIKE DAN AMILASE IKAN LUNJAR (Rasbora lateristriata Blkr.) PADA TEMPERATUR BERBEDA

Ikan lunjar (Rasbora lateristriata Blkr.) tergolong kedalam ikan omnivora. Kemampuan ikan dalam mencerna pakan tergantung pada ketersediaan enzim pencernaan. Aktivitas enzim dipengaruhi oleh kondisi lingkungan, salah satunya adalah temperatur. Penelitian yang berhubungan dengan ikan lunjar (R. lateristriata Blkr.) telah banyak dilakukan, namun kajian aktivitas enzim pencernaan terutama tripsin-like dan amilase terkait dengan perbedaan temperatur masih belum dilakukan. Penelitian ini bertujuan untuk mengetahui aktivitas tripsin-like dan amilase ikan lunjar pada temperatur berbeda dan temperatur optimalnya. Penelitian ini dilakukan secara eksperimental dengan rancangan acak lengkap (RAL) yang terdiri dari 6 perlakuan, yaitu temperatur 20, 30, 40, 50, 60, dan 70ºC. Setiap perlakuan dilakukan pengulangan sebanyak 5 kali. Ikan lunjar yang digunakan adalah ikan dengan panjang tubuh berkisar 4,6±0,2 cm dan berat tubuh berkisar 0,81±0,09 g sebanyak 122 ekor. Konsentrasi tirosin dan maltosa diukur dengan metode spektrofotometer. Data yang diperoleh dianalisis dengan one way analysis of variance (ANOVA) dengan tingkat kesalahan 5% dilanjutkan dengan uji Least Significant Difference (LSD) pada tingkat kesalahan 5%. Hasil penelitian menunjukkan aktivitas tripsin-like ikan lunjar (R. latersitriata Blkr.) tinggi pada temperatur 20-50ºC (0,656±0,26 U.mg-1 protein hingga 1,039±0,232 U.mg-1 protein) dan aktivitasnya rendah pada temperatur 60-70ºC (0,018±0,003 U.mg-1 protein hingga 0,546±0,146 U.mg-1 protein). Aktivitas amilase ikan lunjar tinggi pada temperatur 20-40ºC (3,922±1,061 U.mg-1 protein hingga 8,739±1,805 U.mg-1 protein) dan aktivitasnya rendah pada temperatur 50-70ºC (0,133±0,161 U.mg-1 protein hingga 2,28±1,599 U.mg-1 protein). Aktivitas tripsin-like dan amilase memiliki perbedaan yang signifikan (p<0.05) pada temperatur berbeda.

Yellow rasbora (Rasbora lateristriata Blkr.) is classified in the omnivore. The ability of a fish to digest food depends on the availability of digestive enzymes. Enzyme activity is influenced by environmental conditions, one of which is temperature. Research on yellow rasbora (R. lateristriata Blkr.) has been done a lot. However, studies of enzyme activity mainly trypsin-like and amylase related to temperature differences have not been done. The purpose of this study was to determine trypsin-like and amylase of yellow rasbora at different temperatures and the optimum temperature.The study was conducted experimental, with a completely randomized design (CRD) consisting of 6 treatments, temperatures of 20, 30, 40, 50, 60, and 70ºC. Each treatment was repeated 5 times. In this study, 122 yellow rasbora with a body length of 4.6±0.2 cm and a body weight of 0.81±0.09 g were used. The concentration of tyrosine and maltose was measured by the spectrophotometric method. Data obtained by analysis with one way analysis of variance (ANOVA) with a 5% error rate, continued by Least Significant Difference (LSD) test with a 5% error rate.The results showed that trypsin-like activity of yellow rasbora (R. latersitriata Blkr.) was high at a temperature of 20-50ºC (0,656±0,26 U.mg-1 protein to 1,039±0,232 U.mg-1 protein) and low activity at a temperature of 60- 70ºC (0,018±0,003 U.mg-1 protein to 0,546±0,146 U.mg-1 protein). Amylase activity was high at a temperature of 20-40ºC (3,922±1,061 U.mg-1 protein to 8,739±1,805 U.mg-1 protein) and low activity at a temperature of 50-70ºC (0,133±0,161 U.mg-1 protein to 2,28±1,599 U.mg-1 protein). Trypsin-like and amylase activities of yellow rasbora (R. lateristriata Blkr.) had significant differences (p<0.05) at different temperatures. Based on the results obtained, it can be concluded that trypsin-like and amylase activities of yellow rasbora (R. lateristriata Blkr.) had significant differences at different temperatures. Trypsin-like activity was the same at 20-50ºC and amylase activity increased at 20-30ºC.
