HUBUNGAN KADAR PLUMBUM SERUM DENGAN KADAR HEMOGLOBIN PADA BERBAGAI POLIMORFISMEGEN delta-ALAD : STUDI CROSS SECTIONAL PADA PEKERJA SPBU DI BANYUMAS

Latar Belakang: Timbal dalam polusi udara memiliki dampak negatif pada tubuh. Pekerja SPBU yang terpapar timbal dalam durasi yang lama memiliki risiko mengalami gangguan pada sistem hematopoetik terutama kelainan hemoglobin. Tujuan: Mengetahui hubungan kadar timbal serum dengan kadar hemoglobin pada berbagai polimorfisme gen delta-ALAD petugas SPBU di Banyumas. Metode: Penelitian cross-sectional dilakukan pada 46 pekerja SPBU di Banyumas. Data diambil dalam bulan November-Desember 2011. Darah setiap responden diambil untuk menguji polimorfisme gen delta-ALAD dengan PCR-RFLP, kadar timbal serum dengan menggunakan Spektrofotometer Serapan Atom (SSA) dan kadar hemoglobin dengan menggunakan Hematology Analizer. Uji hipotesis yang dilakukan adalah uji fisher exact. Hasil: Proporsi masing-masing gen delta-ALAD 1-1 (91.3%), 1-2 (8.7% ), dan 2-2 (0%). Pb serum 16.29 ug/dL pada ALAD 1-1 homozigot, dan 20.79 ug/dL pada ALAD 1-2 heterozigot. Hemoglobin pada peserta bervariasi dari 12.20 g/dL sampai 18.20 g/dL dengan rata-rata 14.77 g/dL. Tidak terdapat hubungan (p=0.682) antara timbal serum dan hemoglobin. Tidak terdapat hubungan (p=0.104) antara polimorfisme gen delta-ALAD dan hemoglobin. Kesimpulan: Tidak terdapat hubungan kadar Pb serum dengan kadar Hb pada berbagai polimorfisme gen delta-ALAD pekerja SPBU di Banyumas.

Background: Lead in the air pollution has negative impact to the body. Gas station worker with long duration exposure has risk to have disturbance in hematopoietic system especially hemoglobin disturbance. Aim: To know the association between lead serum levels and hemoglobin level in delta-ALAD gene polymorphism in gas station workers in Banyumas. Metode: This cross-sectional study had 46 gas station workers in Banyumas as the subject. Data was taken between November-Desember 2011. The blood from every subject was taken to identify delta-ALAD gene polymorphism with PCR-RFLP method, lead serum levels with Atomic Absorption Spectrophotometer (AAS) and hemoglobin level with Hematology Analizer. Hypothesis used in this study was fisher exact test. Result: Proportion of each delta-ALAD gene polymorphism were 91.3%, 8.7%, and 0% for ALAD 1-1, ALAD 1-2, and ALAD 2-2 respectively. Lead serum levels were 16.29 ug/dL in ALAD 1-1 homozygote and 20.79 ug/dL in ALAD 1-2 heterozygote. Hemoglobin level in subject was various from 12.20 g/dL to 18.20 g/dL with mean 14.77 g/dL. There was no significant association (p=0.682) between lead serum level dan hemoglobin level. There was no significant association (p=0.104) between delta-ALAD gene polymorphism and hemoglobin. Conclusion: There was no significant association between lead serum level and hemoglobin level in delta-ALAD gene polymorphism in gas station workers in Banyumas.
