KUALITAS SPERMATOZOA DAN STRUKTUR HISTOLOGIS GONAD JANTAN TIGA SPESIES IKAN FAMILI CYPRINIDAE DI SUNGAI BANJARAN

Sungai Banjaran banyak dimanfaatkan dalam bidang perikanan, hal ini dapat diketahui melalui intensnya kegiatan penangkapan ikan di Sungai Banjaran dan luasnya wilayah sungai yang telah di eksplorasi untuk memenuhi kebutuhan perikanan masyarakat sekitar. Para peneliti dan praktisi perikanan mengungkapkan bahwa banyak jenis ikan asli perairan liar terancam punah akibat penangkapan yang tidak terkendali maupun penangkapan dengan menggunakan bahan kimia, hal ini tentu saja akan mengancam keberadaan mereka di alam. Penelitian yang mengkaji perkembangan gonad jantan dan spermatogenesis masih sangat jarang dilakukan. Mengingat pentingnya peran individu ikan jantan pada keberhasilan proses pemijahan di suatu perairan maka status reproduksi individu jantan perlu dikaji secara mendalam. Tujuan penelitian ini yaitu: (1) mengetahui kualitas spermatozoa beberapa spesies ikan tangkapan dari Famili Cyprinidae (2) mengetahui nilai Indeks kematangan gonad (IKG), (3) mengetahui Tingkat Kematangan Gonad (TKG), (4) mengetahui gambaran histologis gonad jantan ikan tangkapan dari Famili Cyprinidae. Manfaat dari penelitian ini adalah memberikan informasi terkait biologi reproduksi Ikan dari Famili Cyprinidae meliputi, kualitas spermatozoa, IKG, TKG, dan untuk mendapatkan gambaran mengenai struktur histologis gonad jantan ikan dari Famili Cyprinidae di Sungai Banjaran pada periode tertentu sebagai dasar penanganan kegiatan domestikasi atau budidaya. Penelitian dilakukan dengan metode Survei, sampel diambil dengan teknik purposive random sampling. Variabel yang diamati dalam penelitian ini yaitu aspek biologi reproduksi tiga spesies ikan jantan dari famili cyprinidae. Parameter yang diukur pada penelitian ini yaitu, kualitas spermatozoa, panjang tubuh ikan, bobot tubuh ikan, bobot gonad jantan, morfologi gonad jantan, dan struktur histologis gonad jantan. Data disajikan dalam bentuk diagram batang. Hasil penelitian menunjukkan bahwa kualitas spermatozoa Ikan Nilem pada periode Maret-Mei yang terbaik adalah pada bulan Mei yakni memiliki rata-rata volume milt sebesar 0,21 ml, berwarna putih, pH 7-8, konsentrasi spermatozoa 2,3454 x109 sel/ml, Motilitas 3, viabilitas 82,55%, frekuensi TKG IV yang tertinggi yakni pada bulan April 86,6%, nilai IKG yang tertinggi terdapat pada bulan April yakni 19,11 %. Pada Ikan Brek jumlah volume milt yang paling tinggi terdapat pada bulan Mei yakni 0,25 ml, dengan milt berwarna putih, dan pH 7-8, konsentrasi spermatozoa tertinggi terdapat pada sampel bulan April 34,465 x109 sel/ml, nilai viabilitas sebesar 88,62%, nilai motilitas senilai 2,9, frekuensi TKG IV tertinggi terdapat pada bulan April yakni 100%, IKG tertinggi yakni 8,2-10% yang dijumpai pada bulan Maret dan Mei sebanyak 3 individu, dan Bulan April sebanyak 1 individu. Kualitas sperma Ikan Lunjar terbaik dijumpai pada bulan Mei yakni memiliki volume milt 0,02 ml, berwarna putih, pH 7-8, konsentrasi spermatozoa 22,435 x109 sel/ml, viabilitas 80%, dan motilitas 3, frekuensi TKG IV tertinggi ditemui pada bulan Mei yakni 70%, IKG tertinggi terdapat pada bulan Mei yakni senilai 8,6%. Struktur histologis gonad jantan pada Ikan Nilem, Brek dan Lunjar memiliki komposisi yang sama, yakni pada ikan yang belum matang gonad komposisi sel spermatogenik yang teramati lebih di-dominasi oleh spermatosit, spermatid, dan spermatogonium, sedangkan ketika telah matang gonad di-dominasi oleh sel spermatozoa dan spermatid.

The Banjaran River is widely used in the field of fisheries, this can be seen through the intensity of fishing activities in the Banjaran River and the wide area of the river that has been explored to meet the fishing needs of the surrounding community. Researchers and practitioners reveal that many species of native fish are threatened with extinction due to uncontrolled fishing or fishing using chemicals, this of course will threaten their existence in nature. Research that examines the development of male gonads and spermatogenesis is still very rarely done. Given the importance of the role of individual fish in the success of the spawning process in waters, the reproductive status of individuals needs to be studied in depth. The aims of this study are: (1) to determine the quality of spermatozoa of several fish species from the family Cyprinidae (2) to determine the value of the gonadal maturity index (GMI), (3) to determine the Gonad Maturity Level (GML), (4) to determine the histological description of the male gonads fish from the family Cyprinidae. The benefit of this research is to provide information related to the reproductive biology of fish from the Cyprinidae family, including spermatozoa quality, GMI, GML, and to get an overview of the histological structure of the male gonads of fish from the Cyprinidae family in the Banjaran River at a certain period as a basis for handling domestication or aquaculture activities. The research was conducted by survey method, the sample was taken by purposive random sampling technique. The variable that being observed in this research is biology reproductive of three male fish from Family Cyprinidae. The parameters measured in this study were fish spermatozoa quality, fish body length, fish body weight, male gonad weight, male gonad morphology, and male gonad histological structure. The data is presented in the form of a bar chart. The results showed that the quality of Nilem Fish spermatozoa in the March-May period was best in May which had an average milt volume of 0.21 ml, white in color, pH 7-8, spermatozoa concentration 2.3454 x109 cell/ml, Motility 3, viability is 82.55%, the highest TKG IV frequency is in April 86.6%, the highest IKG value is in April, which is 19.11%. In Brek Fish the highest volume of milt was found in May, namely 0.25 ml, with white milt, and pH 7-8, the highest concentration of spermatozoa was found in the sample in April of 34,465 x109 cell/ml, viability value of 88.62%, motility was 2.9, the highest TKG IV frequency was in April, namely 100%, the highest IKG was 8.2-10%, which was found in March and May as many as 3 individuals, and April as many as 1 individual. The best quality of Lunjar fish sperm was found in May, which had a milt volume of 0.02 ml, white color, pH 7-8, spermatozoa concentration 22.435 x109 cell/ml, viability 80%, and motility 3, the highest TKG IV frequency was found in in May at 70%, the highest IKG was in May at 8.6%. The histological structure of the male gonads in Nilem, Brek and Lunjar fish have the same composition, namely in immature fish the composition of spermatogenic cells is observed to be dominated by spermatocytes, spermatids, and spermatogonia, whereas when mature, the gonads are dominated by spermatozoa and spermatids.
