PENGARUH METODE THAWING DAN LAMA PENYIMPANAN POST THAWING TERHADAP KUALITAS SPERMATOZOA SAPI FRIESIAN HOLSTEIN (FH)

Penelitian ini bertujuan untuk menganalisis apakah ada interaksi metode thawing dan lama penyimpanan post thawing terhadap motilitas, viabilitas dan abnormalitas semen sapi Friesian Holstein (FH). Materi yang digunakan dalam penelitian yaitu 27 straw semen beku yang berasal dari sapi Friesian Holstein (FH) yang didapatkan dari Dinas Perikanan dan Peternakan Kabupaten Banyumas, eosin nigrosin, dan air. Metode penelitian yang digunakan adalah Rancangan Acak Lengkap (RAL) dengan pola faktorial dengan perlakuan metode dan data Motilitas, Viablitas, Abnormalitas thawing 5oC, 37oC, 27oC masing masing 30 detik dan perlakuan lama penyimpanan post thawing 0 menit, 30 menit dan 60 menit sehingga terdapat 9 kombinasi perlakuan dan diulang 3 kali. Hasil penelitian menunjukkan bahwa metode thawing (A) berpengaruh sangat nyata (P<0,01) terhadap motilitas, viabilitas dan abnormalitas semen sapi Friesian Holstein (FH). Lama waktu penyimpanan post thawing (B) berpengaruh terhadap abnormalitas, motilitas dan viabilitas spermatozoa sapi Friesian Holstein (FH). Hasil analisis rata-rata kualitas spermatozoa sapi Friesian Holstein (FH) pada motilitas yaitu 35,1%, viabilitas 50,78%, dan abnormalitas 15,00%. Metode thawing dan lama penyimpanan post thawing (AXB) berpengaruh sangat nyata (P<0,01) terhadap motilitas, tetapi tidak berpengaruh nyata (P>0,01) terhadap viabilitas dan abnormalitas semen sapi Friesian Holstein (FH). Rataan motilitas, viabilitas dan abnormalitas terbaik yaitu pada metode thawing dengan 37oC dan lama penyimpanan post thawing 0 menit.

The study was to analyze whether there is an interaction between thawing methods and duration of post thawing time on the motility, viability and abnormality of Friesian Holstein semen. The materials used in the study were 27 frozen semen straws from Friesian Holstein obtained from the Banyumas Fisheries and Animal Husbandry Service, eosin nigrosin, and water. The study method used was a Completely Randomized Design (CRD) with a factorial pattern with a thawing temperature 5oC, 37oC, 27oC with duration of all temperature thawing is 30 seconds and duration of post thawing time are 0 minute, 30 minutes, and 60 minutes so there were 9 combinations of treatments and repeated 3 times. The study showed that thawing method (A) had a significant effect (P <0,01) on the motility, viability and abnormality of Friesian Holstein semen. The duration of post thawing (B) had a significant effect (P <0,01) on abnormality, but had no significant effect (P> 0,01) on the motility and viability of semen of Friesian Holstein semen. The results of the analysis of the average sperm quality of Friesian Holstein (FH) cattle on motility were 35.1%, viability was 50.78%, and abnormality was 15.00%. Thawing method and duration of post thawing (AXB) had a significant effect (P <0,01) on motility, but had no significant effect (P> 0,05) on viability and abnormality of Friesian Holstein semen. The best mean motility, viability and abnormality were at 37oC and 0 minute.
