GASTROPROTECTIVE EFFECT OF ZINGIBER OFFICINALE AND ZINGIBER CASUMMUNAR ETHANOL-DYDROCHLORIC ASID INDUCED GASTRIC ULCER IN RAT.

Pendahuluan: Zingiber officinal (ZO) dan Zingiber cassumunar (ZC) memiliki potensi aktivitas gastroprotektif. Dalam penelitian ini, yaitu untuk mengevaluasi efek gastroprotektif dari jahe dan bangle terhadap lesi mukosa lambung yang diinduksi etanol-HCl. Metode: Pada penelitian terdiri dari lima kelompok tikus, masing-masing kelompok terdiri dari 10 ekor tikus. Kelompok I (kelompok kontrol negatif) diberikan tween 80 (1%), kelompok II (kontrol positif) diberikan Lansoprazol (30 mg/kgBB) per oral. Kelompok III ekstrak Zingiber officinale (EZO) 200 mg/kg BB, kelompok IV ekstrak Zingiber cassumuna (EZC) 400 mg/kg BB, kelompok V kombinasi ekstrak etanol Zingiber officinale dan Zingiber cassumuna (CEZOZC) 100:200 mg/kg BB Semua kelompok diberi perlakuan selama 6 hari. Pada hari ke-7, semua kelompok diinduksi dengan etanol-HCl (1:1) 0,2 ml/25 gBB. Empat jam kemudian tikus dikorbankan dan dibedah lambung nya. Perhitungan leukosit, panjang lesi, histologi (pewarnaan H & E), indeks ulkus dinilai untuk menentukan potensi ekstrak dalam pencegahan ulkus lambung. Hasil: Profil histopatologi menunjukkan perbaikan ulkus dan penurunan infiltrasi sel inflamasi neutrofil pada kelompok perlakuan. Terjadi peningkatan jumlah leukosit, skor lesi, indeks ulkus, dan rasio kuratif. Namun, tidak ada perbedaan yang signifikan antara ekstrak tunggal dan kombinasi. Kesimpulan: Z.officinale, Z. cassumun, dan ekstrak kombinasi memiliki efek gastroprotektif pada tikus. Kata kunci: Zingiber officinale, Zingiber cassumunar, gastroprotektif, tikus.

Introduction: Zingiber officinal (ZO) and Zingiber cassumunar (ZC) have the potential gastroprotective activity. In this study, we evaluated the gastroprotective effects of C. longa and A. galanga extract against gastric mucosal lesions induced by ethanol-hydrochloric acid (HCl) in rat model. Method: The experimental design comprised five groups of rats, each group consist of 10 rats. Group I (normal control group) received tween 80 (1%), group II (positive control) was administered Lansoprazole (30 mg/kgBW) orally. group III extract Zingiber officinal (EZO) 200 mg/kg BB, group IV extract Zingiber cassumuna 400 mg/kg BB, group V combination of ethanol extract Zingiber officinal and Zingiber cassumuna (CEZOZC) 100:200 mg/kg BW All groups were treated for 6 days. On the 7th day, all groups were induced by ethanol-HCl (1: 1) 0.2 ml/25 gBW. Four hours later the rat was sacrificed and dissected for gastric. Gross ulcer urea, lesion length, histology (H & E staining), ulcer-index were assessed to determine the potentiality of extract in prevention against gastric ulcers. Results: Histopathological profile showed ulcer repair and reduced neutrophil inflammatory cell infiltration in the treatment group. There was also an increase in the leukocyte count, lesion score, ulcer index, and curative ratio. However, there was no significant difference between single and combined extracts. Conclusions: Z.officinale, Z. cassumun, and combination extract have gastroprotective effect in rat model. Keywords: Zingiber officinal, Zingiber cassumun, gastroprotective, rats
