The Effect of Extraction Time and Temperature Combination on Yield of β-glucan From Mycelium and Fruit Bodies of Auricularia auricula

The main objectives of this research were to determine the effect of extraction time and temperature treatment and to determine the most optimum treatment on yield of β-glucan from mycelium and fruit bodies of Auricularia auricula. The research was conducted using experimental method of Completely Randomized Design (CRD) with three times repetitions. The treatments applied were variations of different extraction time and temperature, which carried out in water bath for treatments of 3 hours at 100oC (T1), 4 hours at 100oC (T2), 5 hours at 100oC (T3), and also carried out in the autoclave for treatments of 15 minutes at 121oC (T4), 30 minutes at 121oC (T5), and 45 minutes at 121oC (T6). The independent variable was the extraction time and temperature treatment, while the dependent variable was the yield of β-glucan. The main parameter was the weight of β-glucan and the supporting parameter was the final pH medium. Data analysis was performed by Analysis of Variance (ANOVA) with 95% accuracy level, followed by the Duncan’s Multiple Range Test (DMRT). The result showed that the combination of extraction time and temperature significantly affected the β-glucan yield of Auricularia auricula. The most optimum treatment for extraction using waterbath was 5 hours extraction time and 100oC temperature, meanwhile the most optimum treatment for extraction using autoclave was 15 minutes extraction time and 121oC temperature.
