Latar Belakang: Diabetes melitus merupakan salah satu penyebab dari berbagai komplikasi penyakit yang ditandai dengan hipeglikemia, keadaan hiperglikemia menyebabkan penurunan kadar glutation. Ekstrak daun Tihtonia diversifolia merupakan ramuan herbal yang digunakan pada masyarakat untuk penyakit diabetes melitus dan secara ilmiah mampu menurunkan kadar glukosa darah.
Tujuan: Tujuan penelitian ini adalah untuk mengetahui efek perlindungan ekstrak daun Tithonia diversifolia dalam meningkatkan kadar glutation pada Tikus Sprague Dawley yang diinduksi nicotinamide-streptozotocin.
Metode penelitian: Penelitian ini merupakan penelitian eksperimental dengan post test only with control group design. Dua puluh satu ekor tikus putih dibagi dalam 5 kelompok. Kelompok 1: kontrol sehat (SO, n=4), kelompok 2: kontrol sakit (SN, n=4), kelompok 3 (n=4), kelompok sakit dan diberikan ekstrak etanol daun kembang bulan 25 mg/kgBB selama 28 hari, kelompok 4 (n=4), kelompok sakit dan diberikan ekstrak etanol daun kembang bulan 50 mg/kgBB selama 28 hari dan kelompok 5 (n=5) adalah kelompok sakit dan diberikan ekstrak etanol daun kembang bulan 100 mg/kgBB selama 28 hari. Kadar glutation diukur setelah proses 28 hari selesai dengan mengambil spesimen serum tikus dan kemudian diukur menggunakan ELISA dalam satuan ukuran mg/dl.
Hasil: Rerata Kadar glutation pada kelompok 1,2,3,4 dan 5 masing-masing adalah 207.18±60.20; 135.10±10.15; 143.68±24.27; 160.78±29.50; dan 179.84±32.82 mg/dL. Hasil uji One Way ANOVA hemoglobin menunjukkan nilai p=0,065(p>0,05). 
Kesimpulan: Ekstrak daun Tithonia diversifolia dengan berbagai dosis tidak dapat meningkatkan kadar glutation pada tikus diabetes.

Background: Diabetes mellitus is one of the causes of various complications of diseases that date with hypeglycemia, hyperglycemia causes a decrease in glutathione levels.. Tihtonia diversifolia leaf extract is an herbal formulation used in the community for diabetes mellitus and is scientifically able to reduce blood glucose levels.
Purpose: The aim of this study was to determine the protective effect of Tithonia diversifolia leaf extract in increasing glutathione levels in Sprague Dawley rats induced by nicotinamide-streptozotocin.
Research method: This research is an experimental research with post test only with control group design. Twenty one white rats were divided into 5 groups. Group 1: healthy control (SO, n = 4), group 2: sick control (SN, n = 4), group 3 (n = 4), sick group and given ethanol extract of flower leaves of 25 mg / kg BW for 28 days , group 4 (n = 4), the sick group was given ethanol extract of leaves of the moon 50 mg / kgB flower for 28 days and group 5 (n = 5) was the sick group and was given ethanol extract of flower leaves of 100 mg / kgBW for 28 days .
Results: Average Glutathione levels in groups 1,2,3,4 and 5 were 207.18 ± 60.20, respectively; 135.10 ± 10.15; 143.68 ± 24.27; 160.78 ± 29.49; and 179.84 ± 32.82 mg / dL. The results of the One Way ANOVA hemoglobin test showed p value = 0.065 (p> 0.05).
Conclusion: Tithonia diversifolia leaf extract at various doses could not increase glutathione levels in diabetic rats.
