Gonadal Development of Osteochilus vittatus Exposed to Potassium Dichromate during Embryonic and Post-Embryonic Period

Potassium dichromate, K2Cr2O7, is a common inorganic chemical reagent, commonly used as an oxidizing agent in industrial applications. As with all hexavalent chromium compounds, it is acutely and chronically harmful to health. Prolonged exposure of hexavalent chromium can induce adverse effects in water biota, especially fish, such as damage to DNA and tissue structures, behavioral modifications, lifespan, metabolic changes, osmoregulation disrupt, and survival and growth rate. An experimental research has been conducted in Osteochilus vittatus, to evaluate the effect of K2Cr2O7 on primordial germ cells (PGCs) and the expression of vasa and to determine the tolerable concentration of this substance to Osteochilus vittatus gonadal development. Day 1 post-hatching larvae (1-dph), 1-month post-larvae, 2, and 3 months juvenile were exposed to potassium dichromate for one month. The vasa expression was determined using qPCR. The PGCs and gonadal development were evaluated from paraffin-embedded section, stained with Haematoxylin-Eosin. The result of this research report for the first time, partial sequence of vasa and its expression from the first four months post-hatching Osteochilus vittatus. The vasa expression was measured as ΔCT of the qPCR product, ranging from -3.0065 to 2.944. The exposure of K2Cr2O7 for 1 month to post-hatching larvae, 1-month, 2-months, and 3-months juvenile lead to the decrease of PGCs number and the delay of gonadal development. There was a consistent pattern on the effect of the toxic at all tested developmental stages, in which the higher concentration resulted in the lower PGCs number. The number of PGCs in juvenile exposed to 2.5 and 5 ppm K2Cr2O7 was significantly lower (p<0.01) than control. This research summarized the adverse effects that K2Cr2O7 cause to Osteochilus vittatus gonadal development. A meager exposure of only 2.5 ppm had already disrupt the expression of vasa, extremely reducing PGCs number, and damaging the whole gonad beyond repair.
