PENGARUH LOGAM BERAT Cd TERHADAP EKSPRESI GEN cGnRH PADA IKAN NILEM (Osteochilus hasselti C.V) BETINA

Kadmium (Cd) adalah logam berat yang memiliki efek toksik baik pada konsentrasi tinggi maupun rendah. Kadmium dapat menyebabkan efek letal yaitu kematian maupun sub letal yang menyebabkan gangguan berbagai fungsi organ termasuk organ reproduksi. Dampak kadmium terhadap ikan dapat menghambat sistem reproduksi dan ekspresi gen cGnRH. Tujuan dari penelitian ini adalah untuk mengetahui pengaruh logam kadmium terhadap ekspresi gen cGnRH dan konsentrasi yang dapat menganggu ekspresi gen cGnRH pada ikan nilem (Osteochilus hasselti) betina. Metode yang digunakan adalah metode eksperimental, dengan desain penelitian Rancangan Acak Lengkap (RAL) Penelitian dilakukan melalui 4 tahapan pengambilan sampel yang terdiri dari pengambilan hipotala

Cadmium (Cd) is a heavy metal that has toxic effects at both low and high concentrations. Cadmium can cause lethal effects, namely death or sub-lethal which causes disruption of various organ functions including reproductive organs. The impact of cadmium on fish can inhibit the reproductive system and expression of the cGnRH gene. The purpose of this study was to determine the effect of cadmium metal on cGnRH gene expression and the concentration that can interfere with the expression of the cGnRH gene in female nilem (Osteochilus hasselti) fish. The method used was an experimental method, with a randomized design research (CRD) method. The research was carried out through 4 stages of sampling consisting of hypothalamus collection, RNA isolation, DNAse treatment, measurement of RNA concentration, and Real-Time PCR. Samples were given 4 different heavy metal treatments for 4 weeks with sampling every 2 weeks. The results showed that the concentration value of the female nilem sGnRh gene expression ranged from 0,0652 – 1,1642 and showed no significant difference (P> 0.05) between all treatments given heavy metals to the control treatment. This can be due to the short exposure time, cadmium concentration, and the adaptability of the fish.
