Kanker rongga mulut merupakan suatu pertumbuhan sel kanker yang terdapat pada mulut, bibir, lidah, palatum, gingiva, dasar mulut, dan mukosa pipi.  Kanker rongga mulut merupakan penyebab kematian tertinggi di Indonesia sebanyak 3-4%. Salah satu cara terapi pengobatan pada sel kanker rongga mulut adalah induksi apoptosis dengan pemberian ekstrak etanol G.lucidum. Pemberian ekstrak etanol G.lucidum diketahui dapat menaikan jumlah apoptosis pada sel kanker payudara (MCF-7), karsinoma paru (95-D) dan karsinoma kolorektal (HT29). Penelitian ini bertujuan untuk mengetahui kemampuan ekstrak etanol G.lucidum terhadap jumlah apoptosis pada sel kanker rongga mulut (KB CCL-17). Kelompok sel KB CCL-17 dibagi menjadi tiga kelompok perlakuan yaitu KP1 (24 μg/ml), KP2 (12 μg/ml), KP3 (12 μg/ml) dan satu kontrol sel (K). Jumlah sel apoptosis dilakukan dengan menghitung persentase sel positif secara metode TUNEL. Hasil dianalisis menggunakan uji One Way ANOVA dan dilanjutkan dengan uji Post Hoc LSD. Jumlah apoptosis pada kultur sel KB CCL-17 masing-masing kelompok yaitu 52,57% (KP1), 32,10% (KP2), 17,74% (KP3) dan 3,45% (K). Ekstrak etanol G.lucidum dapat meningkatkan jumlah apoptosis pada kultur sel KB CCL-17 seiring dengan naiknya konsentrasi. Konsentrasi 24 μg/ml lebih baik dalam meningkatkan jumlah apoptosis dibandingkan dengan konsentrasi 12 dan 6 μg/ml. Terdapat pengaruh pemberian ekstrak etanol G. lucidum terhadap jumlah apoptosis pada kultur sel kanker rongga mulut KB CCL-17.

Oral carcinoma is cancer found in mouth, lips, tongue, palate, gingiva, mouth floor, and cheek mucosa. Oral carcinoma is a common cause of deaths in Indonesia as much 3-4%. One way of theraphy is on cancer cells in oral cavity is treatment using extract of  G. lucidum. A treatment using extract of  G.lucidum is known to increase amount of apoptosis in breast cancer (MCF-7), lung carcinoma (95-D), and colorectal carcinoma (HT29). The aim of this study was to know about the effect of ethanol extract of G.lucidum on amount af apoptosis oral carcinoma (KB CCL-17). The samples were KB CCL-17 cells with three treated groups KP1 (24 μg/ml), KP2 (12 μg/ml), KP3 (24 μg/ml), and one control group (K). Amount of apoptosis was analyzed using TUNEL method by counting the percentage of apptosis positive cells. The data was statistically analyzed using One Way ANOVA and Post Hoc LSD. The results of amount of apoptosis on each groups were 52,57% (KP1), 32,10% (KP2), 17,74% (KP3) and 3,45% (K). Ethanol extract of G.lucidum increased amount of apoptosis in KB CCL-17 cells along with the increase of the dosage. Dosage of 24 μg/ml showed higher increase of amount apoptosis than the dosages of 12 and 6 μg/ml. The conclusion is there is an effect of ethanol extract of G.lucidum on amount of apoptosis in oral carcinoma (KB-CCL17).
