Ayam Kampung mempunyai peran besar bagi kehidupan masyarakat Indonesia terutama di pedesaan. Ayam kampung dapat dikawinkan secara alami maupun buatan. Perkawinan dengan inseminasi buatan diperlukan adanya semen segar dan bisa disimpan lebih lama, penambahan pengencer pada semen yang digunakan saat melakukan IB akan menjadikan volume semen lebih banyak, karena volume semen ayam kampung yaitu berkisar 0,25-0,5 ml/ejakulasi. Metode penelitian yang digunakan yaitu eksperimen dengan menggunakan rancangan acak lengkap dengan pola faktorial yaitu lama penyimpanan (R) r1 = 12 jam, r2 = 24 jam, r3 = 36 jam dan rasio semen: pengencer (P) p0 = 1:0, p1 = 1:5, p2 = 1:10. Materi penelitian yaitu semen dari 12 ekor ayam kampung, mikroskop, serta ringer’s laktat-kuning telur. Hasil penelitian menunjukkan bahwa semakin lama spermatozoa disimpan pada suhu kulkas berpengaruh sangat nyata (P<0,01) terhadap motilitas spermatozoa ayam kampung dan lama penyimpanan spermatozoa berpengaruh nyata (P<0,05) terhadap abnormalitas spermatozoa. Kesimpulan penelitian adalah lama penyimpanan dapat menurunkan motilitas spermatozoa dan meningkatkan abnormalitas spermatozoa dengan mengikuti persamaan regresi Y = 43,595 – 0,404X dan Y = 25,936 + 0,279X. Sebaiknya lama waktu penyimpanan spermatozoa tidak lebih dari 12 jam agar motilitas dan abnormalitas spermatozoa tetap baik dan bisa digunakan untuk pelaksanaan Inseminasi Buatan.

Chicken has a big role for the lives of Indonesian people, especially in rural areas. It can be mated both naturally and artificially. Mated with artificial insemination requires the presence of fresh sperm and can be stored longer, the addition of thinners in sperm used when doing the AI will make the volume of sperm more, because the volume of  chicken sperm ranging from 0.25-0.5 ML/ejaculation. The research method used is an experiment using a completely random design with a factorial pattern i.e. storage time (R) r1 = 12 hours, r2 = 24 hours, r3 = 36 hours and sperm ratio: diluent (P) p0 = 1:0, p1 = 1:5, p2 = 1:10. The research material is the sperm from 12 roosters microscope, and Ringer's lactate-egg yolk. The results showed that the longer the spermatozoa stored at the refrigerator temperature is very noticeable (P < 0.01) on the motility of the kampung rooster spermatozoa and the length of the storage of real effect spermatozoa (P < 0.05) on the abnormality Spermatozoa. The conclusion of the study is the long storage can decrease the motility of the spermatozoa and increase the abnormalities of spermatozoa by following the regression equation Y = 43.595 – 0, 404X and Y = 25.936 + 0, 279X respectivelly. It is better if the length of storage of spermatozoa is not more than 12 hours so that the motility and the abnormalities of the spermatozoa remain good and can be used for the implementation of artificial insemination.
