UJI EMPAT JENIS TEPUNG IKAN TERHADAP KUALITAS METABOLIT SEKUNDER Pseudomonas fluorescens P60 DAN KEMEMPANANNYA TERHADAP PENYAKIT LAYU FUSARIUM TOMAT IN PLANTA

Penelitian bertujuan untuk menguji empat jenis tepung ikan terhadap kualitas metabolit sekunder Pseudomonas fluorescens P60, kemempanannya terhadap penyakit layu Fusarium dan pengaruhnya terhadap pertumbuhan tanaman tomat in planta. Penelitian dilaksanakan di Laboratorium Perlindungan Tanaman dan Screen house Fakultas Pertanian, Universitas Jenderal Soedirman, Purwokerto dari bulan September 2018 – April 2019. Uji in vitro menggunakan Rancangan Acak Lengkap dengan 5 perlakuan dan 5 ulangan, yaitu kontrol, tepung ikan Ursal, Ocialis, Campur Sari Laut, dan produksi PT Anugrah Antari Sejahtera. Uji in planta menggunakan Rancangan Acak Kelompok dengan 5 perlakuan dan 5 ulangan, yaitu kontrol, fungisida (mankozeb 80%), tepung ikan Ursal, produksi PT Anugrah Antari Sejahtera, serta 50% Ursal + 50% produksi PT Anugrah Antari Sejahtera. Variabel yang diamati yaitu zona bening, daya hambat, pertumbuhan perkecambahan, masa inkubasi, intensitas penyakit, selisih tinggi tanaman, panjang akar, bobot segar akar, dan bobot segar tanaman. Hasil penelitian menunjukkan bahwa empat jenis tepung ikan berpengaruh terhadap kualitas metabolit sekunder yang dihasilkan oleh P. fluorescens P60 berdasarkan produksi enzim kitinase dan protease, daya hambat, dan pertumbuhan perkecambahan. Metabolit sekunder P. fluorescens P60 belum mampu mengendalikan penyakit layu Fusarium dan meningkatkan pertumbuhan tanaman tomat, tetapi semua perlakuan dapat meningkatkan kandungan senyawa fenol.

This research aimed to examine four fish meal types on the quality of secondary metabolites derived from Pseudomonas fluorescens P60, its efficacy to tomato Fusarium wilt, and the effect on crop growth in planta. The research was conducted at the Plant Protection Laboratory and the screen house, Faculty of Agriculture, Jenderal Soedirman University, Purwokerto from September 2018 - April 2019. In vitro tests used a completely randomized design with five treatments and five replicates consisted of control, fish meal types of Ursal, Ocialis, Campur Sari Laut, and produced by PT Anugrah Antari Sejahtera. In planta test used a randomized block design with five treatments and five replicates. The treatments were control, fungicide (mancozeb 80%), fish meal types of Ursal, produced by PT Anugrah Antari Sejahtera, and 50% Ursal + 50% produced by PT Anugrah Antari Sejahtera. Variables observed were clear zone, inhibition ability, germination growth, incubation period, disease intensity, crop height difference, root length, root fresh weight, and crop fresh weight. The result of the research showed that four types of the fish meal could affect the quality of secondary metabolites produced by P. fluorescens P60 based on the production of chitinase and protease enzymes, inhibition ability, and germination growth. P. fluorescens P60 secondary metabolites could not control Fusarium wilt and increase the growth of tomato crops, but, all of the treatments could increase phenolic compounds.
