EFEK PROTEKTIF EKSTRAK ETANOL SELEDRI (Apium graveolens L.) TERHADAP KADAR ALBUMIN SERUM TIKUS PUTIH (Sprague dawley) MODEL CHRONIC KIDNEY DISEASE

Latar Belakang: Chronic Kidney Disease (CKD) merupakan kerusakan ginjal dan/atau penurunan laju filtrasi glomerulus (LFG) kurang dari 60 ml/min/1,73m^2 selama minimal 3 bulan yang berakibat pada penurunan kadar albumin serum. Ekstrak etanol seledri (Apium graveolens L.) berfungsi sebagai antioksidan dan antiinflamasi yang berpotensi dapat mencegah kerusakan ginjal sehingga dapat mencegah penurunan kadar albumin. Tujuan: Penelitian ini bertujuan untuk mengetahui efek pemberian ekstrak etanol seledri (Apium graveolens L.) dalam mencegah penurunan kadar albumin serum tikus putih (Sprague dawley) model CKD. Desain Penelitian: Metode penelitian adalah eksperimental dengan post test only with control group design. Dua puluh lima ekor tikus putih dibagi dalam 5 kelompok. Kelompok A (larutan aquades) sebagai sharm operation, kelompok B (larutan aquades) sebagai kontrol sakit, kelompok C (250 mg/kgBB), kelompok D (500 mg/kgBB), dan kelompok E (1000 mg/kgBB). Pada hari ke 15 setelah pemberian ekstrak etanol seledri, kelompok B, C, D, E dibuat model CKD dengan 5/6 sub total nephrectomy. Pemberian ekstrak dilanjutkan sampai hari ke 29. Hasil: Rerata kadar albumin serum kelompok A=3,28±0,084; B=2,90±0,331; C=2,66±0,492; D=2,70±0,291; E=3,16±0,194. Hasil uji Kruskall Wallis rerata kadar albumin serum menunjukkan nilai p=0.021 (p<0,05). Uji post hoc Mann Whitney rerata kadar albumin serum diantara kelompok B, C, D, dan E menunjukan hasil tidak signifikan secara statistik, tetapi pada kelompok E rerata kadar albumin lebih baik dibandingkan kelompok B, C dan D. Kesimpulan: Pemberian ekstrak etanol seledri (Apium graveolens L.) dosis 1000 mg/kgBB bepotensi mencegah penurunan kadar albumin serum tikus model CKD.

Background: Chronic Kidney Disease (CKD) is kidney damage and/or decreased in glomerular filtration rate (LFG) of less than 60 ml/min/1,73m^2 for a minimum of 3 months which results in decreased serum albumin level. Ethanol extract of celery (Apium graveolens L.) functions as an antioxidant and anti-inflammatory which has the potential to prevent kidney damage so that it can prevent a decrease in albumin levels. Objective: This study aimed to determine the effect of celery (Apium graveolens L.) on preventing the decrease of serum albumin levels of rat models (Sprague dawley) CKD models. Research Design: The research method was experimental with post test only with control group design. Twenty five rats models (Sprague dawley) were divided into 5 groups. Group A (aquades solution) as sharm operation, group B (aquades solution) as pain control, group C (250 mg/kgBW), group D (500 mg/kgBW), and group E (1000 mg/kgBW). On the 15th day after administration the ethanol extract of celery, groups B, C, D, E were dissected and made CKD. The extract was continued until the 29th day. Results: The mean result of serum albumin level in group A =3.28±0.084; B=2.90±0.331; C=2.66±0.492; D=2.70±0.291; E=3.16±0.194. Results Kruskall Wallis test serum albumin levels shows a value of p = 0.021 (p <0.05). The Mann Whitney post hoc test mean serum albumin levels between groups B, C, D, and E showed results not statistically significant, but in group E average albumin levels were better than group B, C and D. Conclusion: Administration of ethanol extract of celery ( Apium graveolens L.) can prevent the decrease of rat serum albumin levels in rats models CKD with the most significant dose at dose 1000 mg/kgBW
