APLIKASI BAKTERI ENDOFIT AKAR TOMAT UNTUK MENGENDALIKAN PENYAKIT LAYU BAKTERI PADA TANAMAN TOMAT

Penelitian ini bertujuan mendapatkan bakteri endofit dari akar tanaman tomat di wilayah Kecamatan Baturraden dan mengetahui jenis bakteri endofit akar tomat yang paling efektif dalam mengendalikan penyakit layu bakteri serta pengaruhnya terhadap pertumbuhan tanaman tomat. Penelitian ini dilaksanakan pada bulan November 2017 sampai dengan April 2018 di Laboratorium dan Screenhouse Perlindungan tanaman Fakultas Pertanian Universitas Jenderal Soedirman. Penelitian ini menggunakan Rancangan Acak Lengkap pada pengujian in vitro dan Rancangan Acak Kelompok Lengkap pada pengujian in planta dengan 6 perlakuan dan 4 ulangan. Perlakuan terdiri atas Kontrol (tanpa bakteri endofit) dan 5 isolat bakteri endofit. Variabel yang diamati meliputi zona hambatan, mekanisme daya hambatan, masa inkubasi, intensitas penyakit, AUDPC, tinggi tanaman, jumlah daun, panjang akar, volume akar, bobot segar tanaman, bobot segar akar, bobot kering tanaman dan bobot kering akar. Hasil penelitian menunjukkan bahwa bakteri endofit yang paling efektif mengendalikan penyakit layu bakteri tomat adalah isolat IN4 yang berasal dari desa Karang Tengah mampu menunda masa inkubasi hingga 30 hari setelah inokulasi, efektif menekan intensitas penyakit sebesar 93,90%, mampu menekan nilai AUDPC menjadi 98,76%. Isolat IN4 dari desa Karang Tengah juga mampu meningkatkan variabel pertumbuhan tanaman seperti tinggi tanaman, bobot segar tanaman, bobot segar akar, bobot kering akar dan bobot kering tanaman masing-masing 32,03%, 65, 10%, 78,45% , 22, 47% dan 24, 29%.

The study aimed to obtain endophytic bacteria in the root of tomato plants in Baturraden subdistrict and to observe the types most effective endophytic bacteria from tomato root in controlling bacterial wilt disease and its effect on tomato plant growth. This research was conducted in November 2017 until April 2018 at the Laboratory and Screenhouse of Plant Protection Faculty of Agriculture, University of Jenderal Soedirman. This study used Completely Randomized Design for in vitro test and Group Randomized Design in in planta test with 6 treatments and 4 replications. The treatmens were Controls and 5 type of endophytic bacteria. Observed variables were inhibition zone, mechanism of inhibition, incubation period, disease intensity, AUDPC, plant height, number of leaves, root length, root volume, fresh weight of plant, fresh weight of root, dry weight of plant and dry weight of root. The results showed that the most effective endophytic bacteria in controlling tomato bacterial wilt disease was isolate IN4 from Karang Tengah, this isolate wase to delay incubation period up to 30 days after inoculation, effectively suppress disease intensity of 93.90%, able to reduce the value of AUDPC to 98.76%. Moreover, Isolate IN4 from Karang Tengah treatment was also able to increase plant growth variables such as plant height, fresh weight of plant, fresh root weight, root dry weight and plant dry weight 32,03%, 65,10%, 78.45%, 22, 47% and 24, 29%, respectively.
