PENGARUH ENDOSULFAN TERHADAP EKSPRESI GEN GtH-IIß PADA IKAN NILEM (Osteochilus hasseltii C.V.) BETINA

Endosulfan merupakan neurotoksin yang bersifat persisten, toksisitas tinggi dan termasuk kedalam hydrophobic organic contaminant (HOC). Endosulfan sulit terdegradasi sehingga cenderung meninggalkan residu di lingkungan. Hal tersebut berdampak pada kelangsungan hidup biota air termasuk ikan karena menghambat reproduksi ikan. Tujuan penelitian ini yaitu untuk mengetahui pengaruh konsentrasi dan waktu paparan endosulfan yang mengganggu ekspresi gen GtH-IIß ikan nilem (Osteochilus hasseltii C.V.) betina. Metode penelitian menggunakan metode eksperimental dengan desain penelitian Rancangan Acak Lengkap (RAL). Perlakuan paparan endosulfan yaitu sebesar 0 ppb; 0,88 ppb; 1,76 ppb dan 2,64 ppb. Waktu paparan dilakukan selama empat minggu dengan interval pengambilan sampel setiap dua minggu. Penelitian dilakukan melalui 4 tahap yaitu pengambilan hipotalamus, isolasi RNA, DNAse Treatment dan pengukuran ekspresi gen GtH-IIß menggunakan Real-Time PCR. Data kuantitatif nilai ekspresi gen GtH-IIß dianalisis dengan one way anova dan uji BNT. Hasil penelitian menunjukkan bahwa paparan endosulfan dengan konsentrasi dan waktu paparan yang berbeda tidak berpengaruh nyata terhadap nilai ekspresi gen GtH-IIß ikan nilem betina.

Endosulfan is a neurotoxin persistent, high toxicity and belongs to the hydrophobic organic contaminant (HOC). Endosulfan is difficult to degrade so it tends to leave a residue in the environment. This has an impact on the survival of aquatic biota including fish because it inhibits the reproduction of fish. The purpose of this study was to determine the effect of concentration and exposure time of endosulfan that can interfere with GtH-IIß gene expression in Osteochilus hasseltii C.V. females. The method used experimental method with Completely Randomized Design. The treatment of endosulfan exposure was 0 ppb; 0.88 ppb; 1.76 ppb and 2.64 ppb. The exposure time is carried out for four weeks with a sampling interval every two weeks. The study was conducted through 4 stages of hypothalamus removal, RNA isolation, DNAse Treatment and GtH-IIß gene expression measurements using Real-Time PCR. The quantitative data in the form of GtH-IIß gene expression were analyzed by one-way-anova and BNT test. The results showed that endosulfan exposure with different concentration and time did not significantly affect the GtH-IIß gene expression in nilem females.
