KAJIAN Trichoderma harzianum ISOLAT JAHE DALAM FORMULA CAIR BERBAHAN TEPUNG JAGUNG TERHADAP PENYAKIT REBAH SEMAI (Pythium sp.) PADA BIBIT MENTIMUN

Penyakit rebah semai yang disebabkan Pythium sp. merupakan salah satu penyakit utama yang menurunkan produksi mentimun. Penggunaan pestisida sintetis menimbulkan potensi bahaya bagi kesehatan manusia, degradasi lingkungan, hilangnya keanekaragaman hayati, dan perubahan ekosistem jika tidak ditangani secara tepat. Trichoderma harzianum adalah agensia mapan dan mudah didapatkan sebagai alternatif pengendalian patogen tanaman. Kendala yang dihadapi dalam penerapan mikroba ini adalah medium pembawanya. Tepung jagung merupakan salah satu medium alternatif yang dapat digunakan untuk formula medium cair T. harzianum. Penelitian ini bertujuan untuk mengetahui konsentrasi tepung jagung yang tepat untuk medium cair T. harzianum, mengetahui pengaruh aplikasi T. harzianum dalam berbagai konsentrasi medium cair tepung jagung terhadap penekanan penyakit rebah semai, dan terhadap pertumbuhan bibit mentimun. Penelitian telah dilaksanakan dalam dua tahap, yaitu in vitro di Laboratorium Perlindungan Tanaman dan in planta di rumah plastik Fakultas Pertanian, Universitas Jenderal Soedirman pada bulan September 2017 sampai Januari 2018. Pengujian in vitro menggunakan Rancangan Acak Lengkap dengan 5 perlakuan, meliputi perlakuan formula cair Potato Dextrose Broth (PDB), tepung jagung konsentrasi 5 g/L, 10 g/L, 15 g/L, dan 20 g/L. Masing-masing perlakuan diulang 5 kali. Pengujian in planta menggunakan Rancangan Acak Kelompok dengan 6 perlakuan, meliputi kontrol dan 5 formula T. harzianum. Masing-masing perlakuan diulang 5 kali. Variabel yang diamati meliputi kepadatan konidium, masa inkubasi, kejadian penyakit, area under disease progress curve (AUDPC), potensi tumbuh maksimum, daya kecambah, tinggi tanaman, panjang akar, bobot segar akar, dan bobot segar tanaman. Hasil penelitian menunjukkan, kepadatan konidium T. harzianum tertinggi pada formula medium cair tepung jagung konsentrasi 20 g/L sebesar 3,67x106 konidium/mL, tetapi belum mampu menyamai T. harzianum dalam medium PDB. Aplikasi T. harzianum yang efektif menekan penyakit rebah semai adalah perlakuan T. harzianum dalam formula cair tepung jagung konsentrasi 15 g/L, yaitu mampu menekan kejadian penyakit 71,43%, menunda masa inkubasi 35,83%, dan nilai AUDPC sebesar 195 %hari.

Damping-off caused by Pythium sp. is one of main cucumber decreasing diseases. The use of synthetic pesticides potential results in hazard to human health, environmental degredation, biological biodiversity loss, and ecosystem changed when the pesticide not handled appropriately. Trichoderma harzianum is settled and easily available agent as plant pathogen control alternative. Problem in the application of this microbe is the carrier media. Corn starch is one of alternative media used for liquid media formula of T. harzianum. This research aimed to determine the right corn starch concentration for T. harzianum liquid media, the effect of the media application on T. harzianum in various concentration of corn starch on suppression of the disease, and on cucumber seedling growth. The research was conducted in two stages, i.e., in vitro test at the Laboratory of Plant Protection and in planta one at the plastic house, Faculty of Agriculture, Jenderal Soedirman University, from September 2017 up to January 2018. Completely Randomized Design was used in in vitro test with five treatments consisted of liquid formula from Potato Dextrose Broth (PDB), from corn starch with concentration of 5 g L-1, 10 g L-1, 15 g L-1, and 20 g L-1. Each treatment was repeated five times. In in planta one, Randomized Block Design was used with six treatments, consisted of control, and five formulas of T. harzianum. Each treatment was repeated five times. Variables observed were conidia density, incubation period, disease incidence, area under disease progress curve (AUDPC), maximum growth potential, germination, plant height, root length, root fresh weight, and crop fresh weight. Result of the research showed that the highest T. harzianum conidia density was found in the formula of corn starch with contentration of 20 g L-1 as 3.67x106 conidia mL-1, but it was not similar to T. harzianum in the PDB. The effective application of T. harzianum to suppress the disease was the liquid with concentration of 15 g L-1, indicated by suppressing the disease incidence as 71.43%, delaying incubation period as 35.83%, and AUDPC as 195 %day.
