Respon Fisiologis Tikus Putih Jantan Model Gagal Ginjal Akut Yang Diinduksi CCl4 (Karbon Tetraklorida) Terhadap Lendir Bekicot (Achatina Fulica)

Penanganan pasien gagal ginjal seperti Haemodialisa dan transplantasi ginjal membutuhkan biaya yang mahal. Penelitian ini bertujuan mengevaluasi respon fisiologis tikus model gagal ginjal akut (GGA) yang diinduksi Karbon Tetraklorida (CCl4) terhadap lendir bekicot (Achatina fulica) berdasarkan kadar Superoxide dismutase (SOD) dan kadar Cu. Sebanyak 42 ekor tikus putih jantan galur Wistar umur 3 bulan dan bobot 200-230 g dengan 7 perlakuan dan 3 kelompok. Kontrol Negatif, Kontrol Toksik, Kontrol Cu (23,3 µg/200gBB), Kontrol Zn (1,6 µg/200gBB), Dosis 1 lendir bekicot (150 µl/200gBB), Dosis 2 lendir bekicot (300 µl/200gBB) dan Dosis 3 lendir bekicot (500 µl/200gBB). Pemberian CCl4 dosis 1,5 ml/kgBB dalam olive oil 1:1 secara intraperitoneal diberikan hanya 1 kali, pada semua perlakuan terkecuali kontrol negatif dan setelah 3 hari diberi lendir bekicot secara oral setiap hari dalam 3 kelompok waktu yaitu 1 minggu, 2 minggu dan 3 minggu sekaligus diambil darahnya. Hasil menunjukkan bahwa tikus induksi CCl4 kemudian diberi lendir bekicot dosis 1, dosis 2 dan dosis 3 mampu menurunkan kadar SOD dan kadar Cu ke level yang sama dengan kadar SOD dikontrol negatif. Hal ini menunjukkan bahwa pemberian lendir bekicot yang mengandung SOD sebagai antioksidan berfungsi menetralisir radikal bebas dari induksi CCl4 yang masuk ke dalam tubuh tikus.

Handling such Haemodialisa patients with kidney failure and kidney transplants are very expensive. This study aimed to evaluate the physiological response of rat models of acute renal failure (ARF) induced by carbon tetrachloride (CCl4) against slime snail (Achatina fulica) based on the levels of Superoxide Dismutase (SOD) and Cu content. A total of 42 rats Wistar male 3 months old and weighs 200-230 g with 7 treatments and 3 groups. Negative control, Control of toxic, Control Cu (23,3 g/200gBB), Control Zn (1,6 g/200gBB), Dose 1 snail slime (150 µl/200gBB)), Dose 2 snail slime (300 µl/200gBB) and Dose 3 snail slime (500 µl/200gBB). Award CCl4 dose of 1,5 ml/kgBB in olive oil 1: 1 intraperitoneally administered only one time, all treatments except the negative control and after 3 days of snail slime was given orally every day in 3 groups of the time is 1 week, 2 weeks and 3 weeks at a time to have blood drawn. The results showed that CCl4 induction mice were then given the snail slime dose 1, dose 2 and dose 3 is able to reduce levels of SOD and Cu levels to levels similar to the levels of SOD negative control. This shows that the administration of snail slime containing SOD function as antioxidants neutralize free radicals from the CCl4 induction into the mouse body.
