APLIKASI METABOLIT SEKUNDER DUA ISOLAT Pseudomonas fluorescens SECARA TUNGGAL DAN GABUNGAN TERHADAP PENYAKIT BUSUK KERING PADA PEPAYA LEPAS PANEN

Penelitian ini bertujuan untuk mengetahui metabolit sekunder isolat Pseudomonas fluorescens terbaik dalam menekan penyakit busuk kering dan pengaruhnya terhadap mutu buah pepaya lepas panen. Penelitian ini dilaksanakan di Laboratorium Perlindungan Tanaman Fakultas Pertanian, Universitas Jenderal Soedirman, Purwokerto, mulai Desember 2015 sampai dengan Februari 2016. Pengujian in vitro, digunakan Rancangan Acak Lengkap (RAL) sedangkan pengujian in vivo menggunakan Rancangan Acak Kelompok (RAK) masing-masing diulang sebanyak enam kali. Perlakuan terdiri atas kontrol, metabolit sekunder P. fluorescens isolat P32, metabolit sekunder P. fluorescens isolat P60, serta metabolit sekunder gabungan isolat P. fluorescens P32 dan P60. Variabel yang diamati yaitu tingkat penghambatan antagonis, masa inkubasi, luas serangan, kadar gula dan tingkat kekerasan dan uji organoleptik. Hasil penelitian menunjukkan bahwa perlakuan dengan menggunakan metabolit sekunder P. fluorescens isolate P60 mampu menekan jamur patogen sebesar 47% secara in vitro, sedangkan pada uji in vivo P. fluorescens isolat gabungan mampu menekan luas serangan sebesar 41%. Hasil uji organoleptik menunjukan semua perlakuan memengaruhi warna, rasa, aroma, tekstur dan kesukaan dengan hasil terbaik pada perlakuan gabungan.

This research aimed to know the best secondary metabolites of Pseudomonas fluorescens isolates toward dry rot and its effect on the quality of postharvest papaya. This research was conducted at the Laboratory of Plant Protection, Faculty of Agriculture, Jenderal Soedirman University, Purwokerto, from December 2015 to February 2016. Used Complete Random Design and Randomized Block Design with six replicates each were used in in vitro and in vivo tests, respectifely the treatments tested were control, secondary metabolite of P. fluorescens P32, secondary of P. fluorescens isolates P60 and secondary metabolite of P. fluorescens P32 and P60 combination. Variables observed were inhibition level, incubation period, attack area, sugar content, hardness level, and organoleptic analyses. Results of the research showed that the best secondary metabolite to suppress the fungal grow in vitro was P. fluorescens P60 as 47%. In in vivo one, the best secondary metabolite was the combination as 41%. The best secondary metabolite from P. fluorescens P60 could decrease the fruit papaya softness as 22%, while the best secondary metabolite could increasing sugar content was P. fluorescens P32 as 18,75%. The treatments affected color, taste, flavor, texture and preffene with the best result was in the combined treatment.
