APLIKASI EKSTRAK MIKROBA LIMA RHIZOSFER BAMBU UNTUK PENGENDALI PENYAKIT BUSUK KERING PEPAYA LEPAS PANEN

Penelitian dilaksanakan di laboratorium perlindungan tanaman Fakultas Pertanian Universitas Jenderal Soedirman mulai Maret 2016 sampai dengan Mei 2016. Penelitian ini dilakukan secara dua tahap, yaitu pengujian secara in vitro dan in vivo. Pengujian secara in vitro dilakukan dengan Rancangan Acak Lengkap. Pengujian secara in vivo dilakukan dengan Rancangan Acak Kelompok. Penelitian ini menggunakan 6 perlakuan dan 4 ulangan. Perlakuannya yaitu: Kontrol tanpa antagonis, ekstrak mikroba rhizosfer bambu tali, wulung, ampel, kasap dan kuning. Variabel yang diamati yaitu tingkat penghambatan antagonis, berat kering miselium, masa inkubasi, luas serangan, tingkat kekerasan dan kadar gula serta uji organoleptik.Hasil penelitian in vitro menunjukkan bahwa perlakuan ekstrak rhizosfer bambu ampel, kuning dan kasap mampu menghambat pertumbuhan jamur Fusarium sp. masing-masing sebesar 24,25, 18,5 dan 13% dan perlakuan ekstrak rhizosfer bambu ampel mampu menekan pertumbuhan miselium sebesar 49,1%. Pada uji in vivo, ekstrak rhizosfer bambu ampel mampu menunda masa inkubasi sebesar 57%, ekstrak rhizosfer bambu kuning dan ampel dapat menekan luas serangan masing-masing sebesar 91,89 dan 86,62%. Perlakuan ekstrak rhizosfer bambu kuning dan ampel positif memengaruhi kekerasan, kadar gula dan warna pada buah pepaya.

This research was conducted at the Laboratory of Plant Protection, Faculty of Agriculture, Jenderal Soedirman University, Purwokerto, from March 2016 until May 2016. This research conducted in two stage, i.e., in vitro and in vivo. Compelitely Randomized Design was used in in vitro and Randomized block design was used in vivo test. This research used six treatments and four replicates. The treatments were control, the microorganisms extracts of tali, wulung, ampel, kasap and yellow bamboos rhizosphere. Variable observed was inhibition level, the dry weight of mycelium, incubation period, attack area, hardness level, sugar content and organoleptic test. Result of the in-vitro research showed that the microorganisms extracts of ampel, yellow and kasap bamboos rhizosphere inhibited the fungus growth as 24,25, 18,5 and 13%, respectively, and the microorganisms extracts of ampel bamboo rhizosphere inhibited the mycelium growth as 49,1%. While in in vivo test the microorganisms extracts of ampel bamboo rhizosphere could delay the incubation period as 57%. The microorganisms extracts of kuning and ampel bamboos rhizosphere could supress the attack area as 91,89 and 86,62%, respectively. The microorganisms extracts of yellow and ampel bamboos rhizosphere positively affected hardness level, sugar content and colour of papaya fruit.
