PENGARUH MUTAGEN SODIUM AZIDE TERHADAP SENSITIVITAS PADI VARIETAS INPARI 31, 32, DAN 33

Penelitian ini bertujuan untuk mengetahui sensitivitas varietas, lama perendaman mutagen sodium azide, pertumbuhan dan fisiologi selama perkecambahan dan pertumbuhan bibit, dan jarak genetik berdasarkan perubahan penampilan varietas padi Inpari 31, 32, dan 33. Percobaan dilaksanakan di Screen House dan Laboratorium Agronomi Fakultas Pertanian universitas Jenderal Soedirman pada bulan Juli - Agustus 2015. Percobaan disusun dalam rancangan petak terpisah yang terdiri atas 15 kombinasi perlakuan dengan tiga ulangan. Varietas padi (Inpari 31, 32, dan 33) ditempatkan sebagai petak utama, dan lama perendaman mutagen sodium azide (0, 5, 10, 15, dan 20 jam) sebagai anak petak. Variabel yang diamati adalah daya berkecambah, kecepatan tumbuh, LD50, tinggi bibit, panjang akar primer, diameter batang, panjang daun, lebar daun, jumlah daun, jumlah daun, warna daun, dan jarak genetik. Hasil penelitian menunjukkan bahwa sensitivitas tertinggi benih yang diuji yaitu varietas Inpari 32 dengan nilai LD50 sebesar 4,96 jam, dan sensitivitas paling rendah yaitu inpari 31 dengan nilai LD50 sebesar 25,81 jam. Inpari 32 menunjukkan perkecambahan dan pertumbuhan bibit yang tinggi dibandingkan Inpari 31 dan 33. Hubungan kekerabatan terdekat terjadi pada varietas Inpari 31 tanpa mutagen dengan Inpari 33 tanpa mutagen, inpari 32 perendaman sodium azide 10 jam dengan Inpari 33 perendaman sodium azide 10 jam, dan inpari 32 perendaman sodium azide 20 jam dengan inpari 33 perendaman sodium azide 20 jam.

This objective of this study was to know to sensitivity in different soaking time of sodium azide mutagen, the germination and seedling growth,and genetic distance based on changes in the morphological characters of some Inpari rice varieties of Inpari 31, 32, and 33. The experiment was conducted at Screen House and Laboratory of Agronomy and Horticulture, Faculty of Agriculture, Jenderal Soedirman University on July to August, 2015. The experiment was conducted by using a split-plot design and consisted of 15 combinations of treatment with 3 replications. Rice varieties (Inpari 31, 32, and 33) were placed as the main plot, and the mutagen of sodium Azide in different of soaking duration (0, 5, 10, 15, and 20 hours) as the subplot. The variables observed were seed germination, growth rate, LD50, seedling height, primary root length, stem diameter, leaf length, leaf width, number of leaf, number of tiller, leaf color, and genetic distance. The result indicated that the highest sensitivity on tested seed was Inpari 32 variety with the LD50 value of 4.96 hours and the lowest sensitivity was Inpari 31 with LD50 value of 25.81 hours. Inpari 32 shows germination and seedling growth were high compared Inpari 31 and 33. The closest phylogenetic relationship occurred between Inpari 31 variety without mutagen and Inpari 33 without mutagen, between inpari 32 with soaking time of sodium azide for 10 hours and Inpari 33 with soaking time of sodium azide for 10 hours, and between inpari 32 with soaking time of sodium azide for 20 hours and inpari 33 with soaking time of sodium azide for 20 hours.
