PERTUMBUHAN MISELIUM Pleurotus ostreatus PADA MEDIUM DAN SUHU YANG BERBEDA UNTUK SEDIAAN BIBIT JAMUR F0 DAN F1

Pleurotus ostreatus sudah banyak dibudidayakan di Indonesia. Faktor keberhasilan budidaya jamur dipengaruhi oleh bibit yang digunakan. Kualitas bibit ditentukan antara lain oleh medium dan suhu. Penelitian ini bertujuan untuk mengetahui: 1) interaksi jenis medium dan suhu yang menghasilkan pertumbuhan miselium P. ostreatus terbaik untuk sediaan F0 dan F1; 2) jenis medium dan suhu pertumbuhan miselium P. ostreatus terbaik untuk sediaan bibit F0 dan F1; dan 3) pengaruh berbagai macam medium dan suhu terhadap pertumbuhan miselium P. ostreatus untuk sediaan bibit F0 dan F1. Penelitian dilaksanakan dalam dua tahap penelitian menggunakan metode eksperimental Rancangan Acak Lengkap pola faktorial. Percobaan pertama terdiri atas dua faktor. Faktor pertama adalah medium (4 taraf), yaitu PDA, SDA, MEA, dan PDYA. Faktor ke dua yaitu suhu (2 taraf), yaitu 24oC dan suhu ruang. Perlakuan diulang 4 kali. Percobaan ke dua terdiri atas dua faktor. Faktor pertama yaitu medium (3 taraf), formula A, formula B, dan formula C. Faktor ke dua berupa suhu (2 taraf), yaitu 24oC dan suhu ruang. Perlakuan diulang 5 kali. Data yang diperoleh dianalisis menggunakan Uji F tingkat kesalahan 5% dan 1%. Hasil penelitian menunjukkan bahwa pada tahap F0, jenis medium, suhu, dan interaksi antara jenis medium dan suhu sangat berpengaruh terhadap diameter akhir koloni miselium dan laju pertumbuhan miselium. Hasil uji BNT menunjukkan bahwa medium PDYA suhu ruang merupakan kombinasi medium dan suhu paling baik untuk pertumbuhan miselium P. ostreatus dengan diameter akhir dan laju pertumbuhan masing-masing yaitu 9 cm dan 0,745 cm/hari. Hasil penelitian tahap F1 menunjukkan bahwa jenis medium dan suhu sangat berpengaruh, sedangkan interaksi antara jenis medium dan suhu berpengaruh terhadap laju pertumbuhan miselium. Hasil uji BNT menunjukkan bahwa medium formula B inkubasi suhu ruang merupakan kombinasi medium dan suhu paling baik untuk pertumbuhan miselium P. ostreatus dengan laju pertumbuhan sebesar 0,5160 cm/hari.

Pleurotus ostreatus have been already cultivated widely in Indonesia. This research has goal to knowing: 1) interaction of medium and temperature which result the best mycelial growth of P. ostreatus for F0 and F1 preparations; 2) medium and temperature which result the best mycelial growth of P. ostreatus for F0 and F1 preparations; and 3) affect of types of medium and temperature to mycelial growth P. ostreatus for F0 and F1 preparations. This research has done in two steps use experimental methode of Completely Randomized Design factorial. The first step contains of two factors. The first factors was type of medium (4 levels), that were PDA, SDA, MEA, and PDYA. The second factor was temperature (2 levels), that were 24oC and room temperature. Each of combined treatment was replicated 4 times. The second step contains of two factors. The first factor is medium (3 levels), that were formula A, formula B, and formula C. The second factor was temperature (2 levels), that were 24oC and room temperature. Each of combined treatment was replicated 5 times. Research data were analyzed using analysis of variance (F test), the results showed that in F0 phase, the type of medium, temperature, and interaction between types of medium and temperature had very significant effects to the average of colony diameter and mycelial growth rate. Results of Least Significant Difference (LSD) test showed that the medium PDYA in room temperature was the best combined treatment for mycelial growth of P. ostreatus with average of colony diameter and mycelial growth rate were 9 cm and 0,745 cm.day-1. The results showed in F1 phase, the types of medium and temperature had very significant affects, and interaction between types of medium and temperature has significant effects to the average of mycelial growth rate. Results of Least Significant Difference (LSD) test showed that the medium formula B in room temperature was the best combined treatment for mycelial growth of P. ostreatus with average of mycelial growth rate 0,5160 cm.day-1.
