Penetapan Kadar Flavonoid Total Dan Formulasi Sediaan Losion Antioksidan Ekstrak Etanolik Daun Alpukat (Persea americana Mill.)

Daun alpukat (Persea americana Mill.) mengandung flavonoid yang memiliki aktivitas sebagai antioksidan. Penelitian ini bertujuan untuk menetapkan kadar flavonoid total, mengetahui pengaruh dari lanolin terhadap sifat fisik dan stabilitas sediaan losion ekstrak etanolik daun alpukat serta persen peredaman formula terbaik terhadap radikal DPPH. Ekstraksi simplisia dilakukan secara maserasi selama 3x24 jam dengan pelarut etanol 70%. Penetapan kadar flavonoid total menggunakan metode kolorimetri. Losion diformulasikan menggunakan basis lanolin dengan variasi kadar 9%, 11%, dan 13%. Sediaan losion yang terbentuk diuji syarat dan stabilitas sifat fisiknya. Formula terbaik yang stabil secara sifat fisik selama penyimpanan diuji aktivitas antioksidannya dengan metode peredaman radikal DPPH. Data bentuk, bau, warna, pH, homogenitas disajikan secara deskriptif. Data viskositas, daya sebar, daya lekat, dan persen peredaman DPPH dianalisis menggunakan ANOVA satu arah dengan taraf kepercayaan 95% dengan uji lanjut LSD untuk viskositas, daya sebar, dan daya lekat serta Dunnett T3 untuk persen peredaman DPPH. Hasil penelitian didapatkan kadar flavonoid total ekstrak daun alpukat sebesar 2,53 ± 0,14% setara dengan kuersetin. Peningkatan kadar lanolin dalam sediaan losion ekstrak etanolik daun alpukat dapat meningkatkan nilai viskositas dan daya lekat tetapi menurunkan nilai daya sebar. Kadar lanolin untuk menghasilkan formula terbaik adalah 11% (Formula 2) dengan viskositas sebesar 2.023-5.047 cP, daya sebar sebesar 6,22-7,27 cm, daya lekat sebesar 1,22-4,52 detik, dan persen peredaman terhadap DPPH sebesar 72,55 %.

Avocado leaves (Persea americana Mill.) containing flavonoids compound that was efficacious as an antioxidant. This study aims to establish the levels of total flavonoids content, to determine the effect of lanolin on the physical properties and stability of ethanolic extract avocado leaf lotion and reduction percent of the best formula against radical DPPH. Simplicia was extracted by maceration during 3x24 hours used 70% ethanol. Determination of total flavonoid content using the colorimetric method. Lotion was formulated using lanolin base concentration 9%, 11%, and 13%. Lotion was tested the stability of physical properties. The best formula that had stable physical properties during storage was tested antioxidant activity by DPPH method. Evaluation of organoleptic, pH, and homogenity were descriptively presented. While the measurement result of viscosity, dispersive power, adhesion and radical scavenging activity were analyzed by one way ANOVA with 95% confidence level with a further test LSD for viscosity, dispersive power, adhesion and Dunnett T3 for radical scavenging activity of DPPH. Results of this study, the total flavonoid content was 2.53 ± 0.14% quercetin in extract. Increased levels of lanolin in lotion formula can increase the viscosity and adhesion but lower the dispersive power. Formula 2 is the best formula which contain 11% lanolin, viscosity obtained in the range 2023-5047 cP, dispersive power obtained in the range 6.22-7.27 cm, adhesion obtained in the range 1.22 to 4.52 seconds, and the radical scavenging activity of DPPH at 72.55%.
