PENGARUH PEMBERIAN EKSTRAK BIJI KURMA (Phoenix dactylifera L.) TERHADAP kADAR LOW DENSITY LIPOPROTEIN (LDL) PADA TIKUS PUTIH MODEL DIET TINGGI LEMAK

Latar Belakang : Penyakit Jantung Koroner (PJK) merupakan salah satu penyebab utama kematian di negara maju dan berkembang yang erat kaitannya dengan kadar LDL. Terapi menurunkan kadar LDL melalui farmakologis cenderung menimbulkan efek samping sehingga perlu terapi lain berbentuk obat tradisional khususnya biji kurma. Biji kurma mengandung pektin, fenolik, kuersetin dan vitamin C yang dapat menurunkan kadar LDL dalam darah. Tujuan : Mengetahui pengaruh pemberian ekstrak biji kurma (Phoenix Dactylifera L) terhadap kadar LDL darah pada tikus putih model diet tinggi lemak. Metode : Penelitian ini adalah penelitian eksperimental dengan desain rancangan acak lengkap dan pendekatan pre and post with control group design. Tiga puluh enam tikus putih galur wistar dengan berat badan ±200g dibagi menjadi 6 kelompok. Kelompok A (kontrol negatif) diberikan Normal Fat Diet (NFD), Kelompok B (kontrol positif) diberikan High Fat Diet (HFD), kelompok C,D dan E diberikan HFD dan ekstrak biji kurma dosis bertingkat dan kelompok F diberikan HFD dan simvastatin. Kadar LDL diperiksa pada awal perlakuan dan akhir perlakuan selama 7 hari. Hasil pemeriksaan dianalisis menggunakan uji analysis of variance (ANOVA) dengan Post Hoc Duncan. Hasil : Hasil rerata selisih kadar LDL kelompok D (25,9 ± 11,1) lebih tinggi dari pada kelima kelompok lainnya. Kelompok E memiliki rerata LDL posttest tertinggi berbeda bemakna terhadap kelompok B (p<0,05). Selisih kadar LDL pre-post kelompok D dan E berbeda bermakna terhadap kelompok A (p<0,05), sedangkan kadar LDL pre-post kelompok C dan F tidak berbeda bermakna terhadap kelompok A (p>0,05). Kesimpulan : Ekstrak biji kurma berpengaruh terhadap penurunan kadar LDL pada tikus model diet tinggi lemak dengan dosis efektif 500 mg/KgBB. Kata Kunci : Ekstrak Biji Kurma, LDL, Simvastatin

Background : Coronary Heart Disease (CHD) is one of main causes of death in eveloped and developing countries that have close relationship with LDL cholesterol levels. Therapy to decrease LDL levels by pharmacological likely can cause side effects, so traditional drug was needed especially date seed. Date seeds contain pectin, phenolic, quersetin and vitamin C that can reduce levels of LDL blood levels. Objective : The purpose of this study was to determine the effect of date seed extract (Phoenix dactylifera L.) on LDL levels in high fat diet rats models. Methods : This study is an experimental research design with a completely randomized design and approach to pre and post with control group design. 36 Wistar strain rats with weight ±200 g were divided into 6 groups. Group A (negative control) was given Normal Fat Diet (NFD), Group B (positive control) was given the High Fat Diet (HFD), group C, D and E were given HFD and date seed extract with 3 levels dose and group F was given HFD and simvastatin. LDL levels checked at the beginning and end of treatment for 7 days. Results test were analyzed using analysis of variance test (ANOVA) with Post Hoc duncan. Results : Delta mean in LDL cholesterol group D (25.9 ± 11.1) was higher than the five other groups. Group E has the highest posttest mean LDL different significant against group B (p <0.05). Delta LDL levels group D and E was significantly to group A (p <0.05), but Delta LDL levels group C and F not significantly to group A (p>0,05). Conclusion : There was an effect of administering date seed extract in lowering high-fat diet rats models LDL levels with effectiveness dose of 500 mg/KgBW Keywords : Date Seed Extract, LDL, Simvastatin
