Ganoderma sp. adalah jamur yang memiliki potensi sebagai jamur obat. Jamur dalam golongan ini dapat dijadikan bahan baku obat herbal karena diketahui mengandung senyawa bioaktif yang memiliki beberapa khasiat, salah satunya sebagai antioksidan. Penelitian tentang potensi Ganoderma sp. isolat lokal belum banyak dilakukan, padahal jamur isolat lokal mempunyai potensi yang baik dan tidak kalah dengan Ganoderma sp. lainnya. Ganoderma sp. isolat Banyumas 4 adalah jamur Ganoderma sp. isolat lokal yang diisolasi dari wilayah Banyumas Jawa Tengah. Penelitian ini bertujuan untuk mengetahui potensi yang dimiliki oleh ekstrak miselium Ganoderma sp. isolat Banyumas 4 sebagai penghasil antioksidan dan mengetahui jenis pelarut yang tepat untuk mendapatkan  kadar antioksidan maksimal Ganoderma sp. isolat Banyumas 4. Metode penelitian yang digunakan dalam penelitian ini adalah  metode eksperimental deskriptif kuantitatif. Data kuantitatif hasil uji DPPH (1,1 Diphenyl-2-picrylhidrazyl) dan total fenol dianalisis menggunakan independent sample T-test, sedangkan data kualitatif hasil uji flavonoid dan terpenoid dideskripsikan guna mengetahui perubahan warna sebelum dan sesudah diberi perlakuan. Hasil penelitian menunjukkan bahwa miselium Ganoderma sp. isolat Banyumas 4 berpotensi sebagai antioksidan walaupun masih digolongkan lemah. Ekstrak dari metanol (polar) mempunyai persentase penangkalan lebih tinggi dibandingkan dengan ekstrak n-heksan (nonpolar) dan memiliki nilai IC50  yaitu 1.252,24 µg/ml serta total fenol 21,19 mg/g.

Ganoderma sp. is a mushroom that have potential as medicinal mushroom. It can be used as raw material for herbal medicine, because it contains bioactive compounds that has multiple properties, such as antioxidant. The research of indigenous isolate Ganoderma sp. hasn’t been widely reported. Ganoderma sp. isolates of Banyumas 4 is Ganoderma sp. local isolates that isolated from Banyumas, Central Java.This research aimed to determine the potential of the mycelium extract of Ganoderma sp. isolates Banyumas 4 as a producer of antioxidant and to determine the extract type of solvent to get the maximum antioxidant levels of Ganoderma sp. isolates Banyumas 4. The method of the research is descriptive quantitative experimental methods. Quantitative data on the test result of DPPH (1,1Diphenyl-2-picrylhidrazyl) and the total of phenol were analyzed using independent T-test sample, meanwhile the qualitative data of the flavonoid and terpenoid test result was described to determine the color change before and after given a treatment. The results showed that the mycelium of Ganoderma sp. Banyumas 4 isolates have potential as an antioxidant, although still classed weak. The extract from methanol (polar) has a higher percentage of deterrence than the n-hexane extract (nonpolar) and the IC50 value is 1252.24 ug/ml and the total phenols 21.19 mg/g.
